摘 要:目的:测定尖吻蝮蛇血凝酶N末端氨基酸序列,并对其止血活性进行分析。方法:采用Edm an法测定尖吻蝮蛇血凝酶N末端氨基酸序列,采用毛细管电泳估算其等电点,根据纤维蛋白出现时间和全血凝时间研究其止血活性。结果:尖吻蝮蛇血凝酶N末端氨基酸序列为VIGGNECDTNEEHFL,等电点为6.59,使用后能明显缩短纤维蛋白出现时间和全血凝时间。结论:尖吻蝮蛇血凝酶具有较强的止血活性。Aim: To determine the N-terminal amino acid sequence of haemocoagulase from Agkistodon acutus venom and study its homeostatic activity. Methods: The N-terminal amino acid sequence of haemocoagulase was determined using Edman assay. Its isoelectric point was estimated by capillary electrophoresis and its hemostatic activity was analyzed based on the times required for fibrin formation and whole blood coagulation. Results: It was found that the N-terminal amino acid sequence of haemocoagulase was VIGGNECDTNEEHFL and that its isoelectric point was 6. 59. The amino acid sequences of haemocoagulases from Agkistodon acutus venom and Bothrope jararaca venom were proven to be of high homology. It could shortened the times required for fibrin forma- tion and whole blood coagulation obviously. Conclusion: The haemocoagulases from Agkistodon acutus venom possess considerable hemostatic activities.
