芪玄益心胶囊对糖尿病大鼠胰腺IL-1β及其mRNA NF-κBp50 IκBа的影响

摘　　要：目的:评价芪玄益心胶囊对糖尿病(DM)大鼠的降糖作用,并探讨其作用机制。方法:以四氧嘧啶腹腔注射致胰腺损伤复制大鼠DM模型,将其随机分美吡达组、美吡达+芪玄益心胶囊高、中、低剂量组、模型组,并设空白对照组。观察各组模型血糖、糖化血清蛋白、胰腺白介素-1β(IL-1β)及其mRNA、核转录因子-κBp50(NF-κBp50)及其抑制蛋白(IκBа)水平,并与空白组对照。结果:模型组及各治疗组与空白组比较,血糖、糖化血清蛋白、IL-1β及其mRNA、NF-κBp50(细胞核)、IκBа(细胞浆)表达均显著增高(P<0.05)。美吡达+芪玄益心胶囊各剂量组与模型组比较,血糖、糖化血清蛋白、IL-1β及其mRNA、NF-κBp50、IκBа表达明显降低(P<0.05)。芪玄益心胶囊与美吡达比较,美吡达+芪玄益心胶囊高剂量组对胰腺IL-1β及其mRNA、NF-κBp50、IκBа改善优于美吡达(P<0.05)。结论:糖尿病大鼠胰腺中存在IL-1β、NF-κBp50的激活,芪玄益心胶囊对其有一定的抑制作用,推测抑制IL-1β-NF-κBp50(IκBа)这一途径的激活,是芪玄益心胶囊降低血糖,胰腺保护作用的重要机制之一。Objective..To evaluate the effect of Capsule Qixuan Yixin on blood sugar and glycosylated hemoglobin in diabet rats,and probe the pharmacological mechanism of Capsule Qixuan Yixin. Methods： Alloxan ip to cause pancreatolysis and duplicate the diabet model of rats. The animals were randomized to Mindiab group, Mindiab companied by Qixuan Yixin capsule of high, middle and low dose group, and control group. Observe blood sugar, glycosylated hemoglobin, expression of mRNA and protein of IL - 1β, NF -kBp50, IKBain the pancreas in all the groups. Results ： Compared with control group,blood sugar, glycosylated hemoglobin, expression of mRNA and protein of IL - 1β, NF -kBp50, IKBa in the pancreas were higher in all placebo group and therapeutic groups. （P 〈 0.05 ）. Compared with placebo group, the rats in Mindiab companied by Qixuanyixin capsule of high, middle and low dose group showed lower level of blood sugar, blood sugar,glycosylated hemoglobin ,expression of mRNA and protein of IL- 1β, NF- kBp50, IKBa. （P 〈 0. 05 ） Compared with Mindiab group,Mindiab companied by Qixuanyixin capsule of high,middle and low dose can refrain those index more effectively than Minidiab do（ P 〈 0. 05 ）. Conclusion：Blood sugar and glycosylated hemoglob increased , IL- 1β and NF - kBp50 were actividted in increased in the pancrease of diabet rats ,which could be refrained by capsule Qixuan Yixin. It is speculated that prevetion of the activiation of IL - 1β and NF - kBp50 is the mechanism of capsule. It is one of the important protective mechanism of Qixuan Yixin Capsule.
