摘  要:目的:研究天然药物榄香烯(Ele)、姜黄素(Cur)、雷公藤内酯醇(Tri)对晶状体上皮细胞(LEC)增殖的抑制作用及细胞信号转导机制,为防治后发性白内障提供实验依据。方法:重组人表皮生长因子(rhEGF)诱导牛LEC增殖;四甲基偶氮唑蓝法(MTT)检测Ele、Cur、Tri对LEC增殖的抑制率;流式细胞术(FCM)检测Ele、Cur、Tri对LEC增殖细胞核抗原(PCNA)的抑制作用;荧光分光光度法检测LEC内[Ca2+]i、放射免疫分析法检测cAMP和cGMP,并分析Ele、Cur、Tri的影响。结果:(1)Ele、Cur、Tri对rhEGF诱导的LEC增殖的抑制率随浓度增加和时间延长而增高(P<0.01);(2)Ele、Cur、Tri明显下调rhEGF诱导增殖的LEC内PCNA蛋白表达,呈量效和时效关系(P<0.01);(3)Ele、Cur、Tri使增殖的LEC内[Ca2+]i和cAMP显著增高(P<0.01)、cGMP显著降低(P<0.01)。结论:天然药物Ele、Cur、Tri均可显著抑制LEC增殖;Ca2+、cAMP和cGMP细胞信号转导是其抑制LEC增殖的重要细胞和分子机制。Ele、Cur、Tri对防治后发性白内障具有广阔的前景。Objective: To investigate the Inhibition of natural medicines, elemene ( Ele), curcumin ( Cur), triptolide (Tri) ,on proliferation induced by recombinant human epidermal growth factor(rhEGF) in lens epithelial cell(LEC) and its mechanism of signal transduction, in order to provide experiment evidence for prevention and treatment of after cata- ract. Methods: Proliferation of cultured bovine LEC were induced by rhEGF; Inhibitory rates of LEC proliferation induced by Ele, Cur, Tri were detected by methyl thiazolyl tetrazolium (MTF) ; Inhibitory effects of expression of proliferating cell nuclear antigen(PCNA) induced by Ele ,Cur,Tri in LEC were assayed via flow cytometer (FCM) ;Concentrations of LEC calcium ([ Ca^2+ ]i )were determined by spectrofluoremeter, intracellular concentrations of cyclic adenosine monophosphate(cAMP) and cyclic guanosine monophosphate(cGMP) of LEC were measured by radioimmunoassay and the influences of Ele, Cur,Tri were analyzed. Results:( 1 ) Inhibitory rates of Ele, Cur,Tri on LEC proliferation induced by rhEGF increased significantly, showing dose-dependent and time-dependent( P 〈 0. 01 );(2)PCNA expression of LEC proliferation induced by rhEGF were down regulated obviously by Ele, Cur,Tri, showing dose-dependent and time-dependent (P 〈 0. 01 ) ;(3 ) Concentrations of[ Ca^2+ ]i and cAMP increased and cGMP decreased significantly in LEC of proliferation in ( hibited by Ele, Cur,rift ( P 〈 0. 01 ). Conclusions: ( 1 ) Natural medicines,Ele, Cur,Tri can inhibit LEC proliferation obviously; (2)Signal transductions of[ Ca^2+ ]i; cAMP, cGMP may be the important cellular and molecular mechanism. (3)There are broad prospect for Ele,Cur,Tri on prevention and treatment of after cataract.
