摘 要:目的:研究姜黄素水解衍生物对体外培养人肝癌细胞HpG2的增殖抑制作用和对肝癌细胞HpG2凋亡调控因子Fas-L、Bcl-2的表达影响。方法:用盐酸和三氯化铝水解姜黄素,获得姜黄素水解衍生物;MTT比色法观察姜黄素水解衍生物对人肝癌细胞HpG2增殖的抑制作用;用免疫组化法检测其对人肝癌细胞HpG2凋亡调控因子基因Fas-L、Bcl-2的表达影响;采用RT-PCR检测c-Myc的表达。结果:姜黄素水解衍生物对人肝癌细胞HpG2有明显的抑制作用(P<0.01),且与姜黄素有显著差异(P<0.001);姜黄素水解衍生物能降低体外培养的人肝癌细胞HpG2的Bcl-2表达,增强Fas-L的表达。能够下调C-Myc的表达。结论:姜黄素水解衍生物有抗肿瘤作用,而且抗肿瘤可能机理可能通过降低肿瘤凋亡调控因子Bcl-2表达,增强Fas-L的表达,下调C-Myc的表达。Objective:To study the effects on the anti - proliferation of ramification of Curcumin hydrolyzed by HCl (RCH) on the H pG2 and the expression of Fas- L, Bcl-2. Methods:Curcumin was hydrolyzed by HCl. MTT was used to observe its effects on anti-proliferation of the HpG2. The Fas -1, Bcl-2 were teseded by immunohistochemical method. The c-myc mRNA was measured by RT- PCR. Resuh:RCH have obvious effects on anti -proliferation of HpG2 cells(P 〈0.01 ). It have significant effects on decreasing Bcl-2 expression and increasing Fas- L expression and down- regu- late the expression of c - Myc. Conclusion. RCH Culd inhihit the growth HpG2 cells, we proposed its mechanisms were probably associated with the decreasing Bcl -2 and increasing Fas - L protein expression and down - regulating the expression of c - Myc.
