高氧致肺纤维化新生大鼠肺组织肾素-血管紧张素系统与Ⅰ型胶原及细胞间黏附分子-1的动态变化

摘  要:目的观察高氧致肺纤维化新生大鼠肺组织血管紧张素转换酶(ACE)、血管紧张素Ⅱ(AngⅡ)、Ⅰ型胶原及细胞间黏附分子-1(ICAM-1)的动态变化。方法足月新生Wistar大鼠120只,随机分为实验组和对照组,各60只。实验组将足月新生Wistar大鼠(连同母鼠)生后即置于有机玻璃氧舱内持续吸入高浓度氧(FiO2:0.9)21d造成高氧肺损伤模型,对照组吸入空气。每组分别于实验后1、3、7、14、21d随机选取6只麻醉后处死。肺组织采用酶联免疫吸附法(ELISA)测定CoⅠ及ICAM-1蛋白含量,用生化分析仪测定ACE的活性,用放射免疫法测定AngⅡ蛋白含量。用逆转录聚合酶链反应(RT-PCR)检测肺组织ACE、AngⅡ及CoⅠmRNA的动态变化并同时观察肺组织形态学变化。结果实验组肺组织各项测量指标在第14天均有明显升高,ACE活性及AngⅡ、Ⅰ型胶原、ICAM-1的蛋白含量分别达到(241.00±31.84)、(707.21±282.90)、(397.97±88.76)和(691.76±90.36)μg/g,除了ACE,其他与对照组比较,差异均有显著性(P〈0.05);而ACE、AreⅡ及Ⅰ型胶原mRNA表达在第14天较对照组均有明显升高,差异均有显著性(P〈0.05);实验组肺组织各项测量指标在第21天达到高峰。ACE活性及AngⅡ、Ⅰ型胶原、ICAM-1的蛋白含量分别达到(249.20±20.19)、(838.22±197.75)、(539.85±81.03)、(577.06±61.17)μg/g,与对照组比较差异均有显著性(P〈0.05);ACE、AngⅡ及Ⅰ型胶原mRNA表达在第21天分别达到2.67±0.52、2.50±0.84及3.0±0.05,与对照组比较差异均有显著性(P〈0.05)。肺组织形态学改变:实验组第14天肺间质细胞增多,肺组织出现纤维化改变;第21天正常肺泡结构消失,残留肺泡直径明显缩小。结论高氧致肺纤维化新生大鼠肺组织ACE、AngⅡ、Ⅰ型胶原、ICAM-1蛋白含量及mRNA�Objective To observe the dynamic changes of renin-angiotensin system, collagen Ⅰ(CoⅠ) and intercellular adhesion molecule 1 (ICAM-1) in lung tissue of neonatal rats with lung fibrosis induced by hyperoxia. Methods One hundred and twenty term neonatal Wistar rats were randomly assigned into model group and air control group. The model group was continuously exposed to hyperoxia (FiO2 = 0.9 )for 21 days, and the air control group was exposed to room air (FiO2 = 0.21). On the 1st, 3rd, 7th, 14th and 21st day of exposure, the subjects were sacrificed. And then, the protein levels of CoⅠ, ICAM-1 were measured by enzyme-linked immunosorbentassay and angiotensin(AngⅡ) by radio-immunity technique, the activity of angiotensin-convertion enzyme(ACE) were measured by biochemistry analysator, and the mRNA expression of ACE, AngⅡ, CoⅠ was measured by RT-polymerase chain reaction. The changes of lung histomorphology were observed. Results The ACE, AngⅡ, CoⅠ and Ⅰ CAM- 1 protein levels of model group were (249.20 ± 20.19) μg/g, (838.22 ± 197.75) μg/g, (539.85 ± 81.03) μg/g and (577.06 ± 61.17)μg/g on the 14th day respectively and increased significantly as compared to the air control group (P〈0.05), and the ACE, AngⅡ, CoⅠ mRNA expression of model group also increased significantly on the 14th day (P〈 0.05). The ACE, AngⅡ, CoⅠ and ICAM-1 protein levels of model group were(249.20 ±20.19) μg/g, (838.22 ± 197.75) μg/g, (539.85± 81.03) μg/g and (577.06± 61.17) μg/g on the 21st day and increased to the peak respectively ( P 〈 0.05), and the ACE, AngⅡ and CoⅠ mRNA expression had the same changes as well. The histopathological examination demonstrated different degrees of fibrosis in the model group. Conclusion The ACE, Ang Ⅱ, Co Ⅰ and ICAM-1 protein levels and their mRNA expression were increased significantly on the 14th and 21st day in lung tissue of neonatal rats with lung fibrosis induced by hyperoxia.
