摘 要:目的:研究人工合成的寡聚脱氧核苷酸(ODN)对人体抗病毒免疫反应的抑制作用,为开发具有免疫调节功能的新型ODN提供筛选平台。方法:将热灭活的人单纯疱疹病毒I型(HSV-1)和流感病毒PR8株(PR8-Flu)作为刺激物,同时以已知的免疫抑制性ODNIRS954作为抑制物,用生物学检测方法分析病毒与ODN共同作用的人外周血单个核细胞(PBMC)培养上清的抗病毒活性,并优化各项实验条件。结果:IRS954能显著抑制HSV-1和PR8-Flu诱导的人PBMC的抗病毒活性。在该实验条件下,自行设计的ODN显示出不同的免疫抑制活性。结论:具有一定序列特点的ODN能够抑制由DNA或RNA病毒诱导的人体抗病毒免疫,该实验方法为研究开发新型免疫调节性ODN奠定了基础。Objective: To investigate the suppressive effect of synthetic oliodeoxynucleotide (ODN) on the anti-viral immune respenses of human and to provide a screening platform for the development of novel ODNs with immunoregulatory activity.Methods: The heat-inactivated HSV-1 and PR8-Flu virus were used as stimulus respectively and the well studied suppressive ODN, IRS954, was selected as suppressor. The human PBMCs were stimulated by the inactivated virus and/or IRS954, then the anti-viral activity of the culture supernatant was tested by the anti-viral bioassay. The experimental conditions were also well optimized. Results: IRS 954 could suppress anti-viral immune responses triggered by HSV-1 and PR8-Hu significantly.In such experimental condition, self-designed ODNs had different suppressive activities. Conclusion: The anti-viral immunity induced by DNA or RNA virus can be suppressed by the ODNs with special sequence, This experimental method sets the foundation for research and development of novel immunoregulatory ODNs.
