摘 要:目的研究壳寡糖减弱乙醇对人胚胎肝细胞(L02细胞)损伤的原因。方法采用MTT法检测壳寡糖减弱乙醇对L02细胞的损伤;结合RT-PCR和Western blot方法分析壳寡糖的保护作用。结果壳寡糖能明显降低乙醇对L02细胞的损伤(P<0.01),尤其是0.2 mg/mL的壳寡糖,而这种作用是通过降低p53基因和蛋白的表达来实现的。结论壳寡糖有保护L02细胞抵制乙醇损伤的作用。Purpose To study the protective effect of oligochitosan on injury induced by ethanol in human embryonic hepatocytes (L02 cells). Methods MTT assay was used for evaluating protection of oligochitosan on L02 cells against ethanol ; RT-PCR and Western blot were used for explaining the reason of protection. Results Oligochitosan notably recovered the injury induced by ethanol ( P 〈 0.01 ), especially the 0.2 mg/mL oligochitosan. The mechanism was possibly that oligochitosan down-regulated p53 expression. Conclusion Oligochitosan could protect human embryonic hepatocytes against the injury induced by ethanol.
