摘  要:目的探讨瘦素(leptin)对大鼠膝关节软骨的外周作用。方法在SD大鼠膝关节腔内注射瘦素100μg后,取胫骨平台,分别用RT-PCR法及免疫组织化学法检测胫骨平台软骨中瘦素及胰岛素样生长因子Ⅰ(IGF-Ⅰ)的变化。结果瘦素无论在RNA水平或蛋白水平都可诱导正常关节软骨IGF-Ⅰ合成。结论瘦素在外周对关节软骨有保护作用,并在骨关节炎的病理生理过程中起到重要的作用。Objective To investigate the peripheral effect of intraarticular injection of leptin on rat cartilage. Methods Leptin(100 μg) was injected into the knee joints of SD rats. Tibial plateaus were collected and processed for the expressions of leptin and insulin like growth factor-Ⅰ (IGF-Ⅰ) by reverse transeriptase-PCR and immunohistochemical analysis. Results Leptin induced the synthesis of IGF-Ⅰ in rat cartilage at both the messenger RNA and the protein levels. Conclusion The findings suggest that leptin, as a key regulator of chondrocyte metabolism, has a peripheral protective function on rat cartilage and may play an important role in the pathophysiology of osteoarthritis.
