摘 要:目的为了研究睾丸网注射法建立转基因小鼠的可行性。方法本试验应用显微注射仪将脂质体包裹的质粒pEGFP-N1通过睾丸网注射到4周龄昆明(KM)小白鼠睾丸生精小管内。注射后6周,与自然发情雌鼠交配,用PCR法检测仔鼠基因组中的外源DNA。结果共注射4只雄性小鼠,3只存活,并且具有交配,受精能力,共获仔鼠63只,其中2只为PCR阳性仔鼠。结论用睾丸网注射法制备转基因动物是可行的。Objective To study the feasibility of establishing transgenic mice carrying enhanced green fluorescent protein gene by means of testis fete injection. Methods Using micropipette plasmid pEGFP-N1 packed by liposome was injected into the testis fete of 4 weeks male KM mice, which were made to mate with spontaneous oestrus female mice at least 6 weeks after the injection. The viral DNA harbored in F1 mice genome was identified by polymerase chain reaction (PCR). Results Among the 4 injected, 3 of them survived and retained their mating ability and fertility. Two of total 63 offspring were detected to have EGFP DNA. Conclusion The data in this study indicated that transgenic animals can be established by testis fete injection.
