工艺控制对复方丹参片抗心肌缺血作用的影响

摘　　要：目的:控制工艺条件制备了有效成分含量不同的复方丹参片,考察其对心肌缺血大鼠T波高度及心率变化的作用。方法:以有效成分含量不同的复方丹参片(工艺参数不同)对大鼠连续灌胃给药3d,末次给药后20min,腹腔注射乌拉坦1g/kg麻醉,测定Ⅱ导联心电图。20min后,正常对照组尾静脉注射等体积生理盐水,其余静脉注射(iv)垂体后叶素(Pit),5s内注完。记录iv Pit前及iv Pit后的心电图变化,并测量T波高度及心率的变化。结果:与模型组比较,工艺A、工艺B三个剂量组、工艺C中剂量在15s时均能显著对抗垂体后叶素引起的T波变化(P<0.05;P<0.01),且工艺B三个剂量组之间存在一定的量效关系,而工艺C低剂量组无此作用;工艺B、工艺C均有缓解垂体后叶素所致的心率减慢的倾向,工艺A无此作用。结论:工艺参数的调整影响复方丹参片有效成分的含量,有效成分需在一定限度以上才对心肌缺血有较稳定的作用。Authors prepared Danshen tablets under different process control plans, in an attempt to observe their effects on T-wave values and heart rates in myocardial ischemia rats. In the study, rats were given orally Danshen tablets prepared under different control plans for 3 days. 20 min after the last intragastric administration, the rats were injected with Urethane 1g·kg-1 intraperitoneally to anesthetize, and the ECG was measured （lead Ⅱ ）. 20 min later, the normal control group was injected with normal saline of an equal volume on vena caudalis, and other groups were injected with pituitrin （Pit） intravenously in 5 s. Changes of ECG T waves and heart rates before and after intravenous（iv） injection of pituitrin were recorded. Comparing with the model group, process A, three different dosage groups of process B, and the medium dosage group of process C saw an inhibited elevation of T-wave as the result of intravenous injection of pituitrin at 15 s（P〈0.05;P〈0.01）. Three different dosage groups of process B exhibited a quantity/effect relation, with the lower dosage group under process C showing no effects. Both processes B and C appeared to relieve the heart rate decrease caused by intravenous injection of pituitrin. Process A saw no such an effect. Authors believe that the preparation process can be fine tuned to allow the tablets containing needed effective constituents for a best possible anti-myocardial ischemia effect.
