中药舒筋止痛液对软骨细胞凋亡状态的影响

摘　　要：目的:观察中药舒筋止痛液对软骨细胞凋亡状态的影响。方法:从幼兔关节中获取软骨细胞;通过检测不同浓度中药培养软骨细胞的MTT值,筛选中药浓度;NO诱导软骨细胞凋亡后,将中药分为高、中、低浓度组,配以阳性对照组和阴性对照组分别对凋亡软骨细胞进行干预培养;采用形态学观察、流式细胞检测等方法评价中药对软骨细胞凋亡状态的影响。结果:中药中浓度组干预的软骨细胞其MTT值高于其他各组,细胞凋亡状态恢复较好,流式检测显示其凋亡率和阳性对照组无显著性差别,低于其他组别。结论:中药舒筋止痛液临床治疗骨关节炎有效与抑制软骨细胞的凋亡状态相关。Objective To observe the effect of Shujinzhitong（舒筋止痛） liquid on apoptotic chondrocyte in vitro. Methods Chondrocytes from the joint of young rabbit were acquired and cultured to select the effective dose through assaying the MTF of chondrocyte cultured with different concentrations of Shujinzhitong liquid; thus to induce chondrocyte apoptosis with nitrogen monoxide, then separately cultured by five groups： in Group A, the final concentration of Shu- jinzhitong liquid was 3.6 mg/mL; in Group B, the final concentration of Shujingzhitong liquid was 0.6 mg/mL; in Grooup C, the final concentration of Shujinzhitong liquid was 0.36 mg/mL; in Group D, positive control, transforming growth factor β1, the final concentration was 10 ng/mL; in Group E, negative control, DMEM with 5% fetal bovine serum. The morphologic changes of chondrocyte were observed by inverted microscopy and apoptosis was investigated by flow cytometry. Results The group of 0.6 mg/mL Zhujinzhitong liquid was better than other groups assayed by MTF; the Group B could significantly resume the apoptotic chondrocyte in morphology, the apoptosis rate of Group B and positive control is lower than other groups. Conclusion Shujinzhitong liquid can effectively inhibit chodroocyte apoptosis in vitro, it may be the basis for clinical treatment of osteoarthritis.
