摘  要:斯氏假单胞菌(Pseudomonas stutzeri)A1501是一株分离自中国南方水稻根际土壤的联合固氮菌。该菌在无氨和微好氧条件下可将空气中的氮气转化为植物可以直接利用的铵。A1501基因组测定工作已经完成,根据A1501菌基因组的功能注释对该菌的中心代谢、能量合成及环境适应性等方面进行了生物信息学分析。结果发现,A1501菌的物质代谢途径具有多样性,除不能利用EMP途径代谢己糖外,该基因组含有几乎所有编码Entner-Doudorff途径(ED)、磷酸戊糖途径(HMP)、糖酵解途径(EMP)、三羧酸循环(TCA)以及乙醛酸途径中关键酶类的基因。此外,基因组分析鉴定了252个与能量产生相关以及3套编码电子传递复合体(1个Nqr和2个Rnf复合体)的基因簇。为进一步研究A1501菌的碳代谢调控及C-N偶联机制提供了重要的理论依据。Pseudomonas stutzeri A1501,isolated from rice paddy soil in South-China,can be able to fixe nitrogen under microaerobic conditions in free-living state. The ammonia,production of nitrogen fixation,can be directly absorbed by host rice. Genome sequencing of A1501 was completed,which brings the chance to investigate mechanisms of fixation nitrogen,material metabolism and energy conversion. Biuinformatics analysis of A1501 genome sequence on central metabolic pathways,energy production and environmental adaptation was performed. The results implied that A1501 has diversity metabolic pathways. Genes involved in the complete Entner-Doudorofff pathway,pentose phosphate pathway and tricarboxylic acid cycle were identified. A1501 strain cannot use Embden-Meyerhof-Parnas pathway for hexose utilization owing to absence of 6-phosphofructokinase. A1501 encodes 252 genes that are being responsible for energy production and conversion. One set of nqr and two sets of rnf cluster encoding for putative electron transport complex,was identified in A1501. Analysis of metabolic and energy production pathways will provide theoretical basis for the improvement of fixation nitrogen efficiency and studies on carbon-nitrogen regulatory mechanism.
