摘  要:目的:建立肝宁颗粒(大黄、川芎)中大黄的含量测定方法。方法:用HPLC法测定大黄中的5种蒽醌苷元。选用NucleosilODS柱,以甲醇-0.1%磷酸溶液(85:15)为流动相,检测波长430nm。结果:HPLC法5种葸醌苷元的线性范围为芦荟大黄素0.0416~0.2912txg,大黄酸0.0312~0.2598μg,大黄素0.0458~0.3203μg,大黄酚0.0496~0.3472μg,大黄素甲醚0.0206~0.1448μg;5种蒽醌苷元的平均回收率为均在96.6%~98.4%。结论:方法简便,准确,灵敏度高,准确性强,重现性好,建立的方法可用于肝宁颗粒的质量控制。AIM: To determine Radix et Rhizoma Rhici in Ganning Granula(Radix et Rhizoma Rhei, Rhizoma Chuanxiong). METHODS: In order to determine five anthraquinone aglycones of Radix et Rhizoma Rhei, the HPLC system consisted of Nucleosil ODS column, methand-0. 1% phosphoric acid(85: 15) mixture as mobile phase, with detection wavelength at 430 nm. RESULTS: HPLC showed that the linearity ranges were as follows:aloe-emodin concentration was at the range of 0. 041 6-0. 291 2μg, the rhein concentration was at the range of0. 037 1-0.259 8 μg, the emodin concentration was at the range of 0. 045 8-0.320 3 μg, the chrysophanol concentra-tion was at the range of 0. 049 6-0. 347 2 μg, the physcion concentration was at the range of 0.020 6-0. 144 5 μg. The average recoveries of five anthraquinone aglycones were between 96.6% -98.4%. CONCLUSION : The meth-od is simple, accurate, sensible and with good repeatability and can be used for quality control of Ganning Granule.
