摘 要:目的验证酶联免疫吸附(EUSA)法定量测定生物样品中的鼠神经生长因子(mNGF)。方法利用双抗体夹心原理,建立mNGF测定方法学,考察方法学特异性、灵敏度、线性范围、精密度与准确度;并用此法研究比格犬肌注mNGF后的药物动力学。结果方法特异性良好,线性范围31.2~2000Pg·mL^-1,可靠定量下限(LLOQ)为31.2pg·mL^-1批内及批间精密度均〈16%,准确度为-9.3%~12.1%。结论ELISA法灵敏度高、通量大、重复性、准确度及精密度好。适用于生物样品中mNGF的定量检测。Objective To develop and validate an enzyme linked imrnunosorbent assay (ELISA) method for the quantification of mouse nerve growth factor (mNGF). Methods The specificity, accuracy, precision and sensitivity of this method were determined. The method was applied in a pharmacokinetic study in beagle dogs treated with a single dose of mNGF. Results Both the standard samples provided by the kit and mNGF appeared typical standard curve within 31.2- 2 000 pg·mL^-1. The coefficient variation of intro- and intra batch was within 16%. The accuracy was -9.3%- 12. 1%. The lower limit of quantitation (LLOQ) was 31.2 pg · mL^-1. Conclusion The present ELISA method is precise, sensitive and reproducible for the quantitation of mNGF.
