摘 要:目的采用快速心房起搏致犬急性心房颤动(简称房颤)模型,观察心房肌缝隙连接蛋白40和43(Cx40、Cx43)含量的改变以及胺碘酮干预的效果。方法18只成年杂种犬,随机等分为三组,即正常对照组、急性房颤组、胺碘酮组。快速心房起搏,对照组不起搏,胺碘酮组先静脉注射胺碘酮负荷量3mg/kg,10~15min内注入,后以维持量1.5mg/kg静脉滴注进行干预,另两组给予等容量的生理盐水。连续刺激并且房颤8h后,取右心耳组织,用Westernblot检测Cx40和Cx43的含量,用LuciferYellow划痕标记荧光传输技术在荧光显微镜上观察细胞缝隙连接通讯状态。结果急性房颤组心房肌组织Cx40和Cx43含量较对照组降低(P均<0.05),胺碘酮组Cx40、Cx43含量较正常对照组低,差异有显著性(P均<0.05),较房颤组含量差异无显著性(P>0.05),胺碘酮可以改善急性房颤时细胞缝隙连接通讯的传导。结论急性房颤犬缝隙连接蛋白40和43发生了结构重构,从而影响细胞间通讯,胺碘酮可以改善细胞间电信号传导,但不能改善缝隙连接结构重构。Objectve To investigate the changes of connexin (Cx) 40 and 43 levels and the effects of amiodarone on them in dog with atrial fibrillation induced by rapid pacing. Methods 18 adult mongrels were randomly and equally divided into 3 groups ( n = 6 ) : normal control group, acute atrial fibrillation group, amiodarone group. Atrial pacing was undergone at the frequency of 600 times per minute for 8 hours in the latter two groups. The infusion of amiodarone with 3 mg/kg for loading dose within 10 15 min and then 1.5 mg/kg for maintenance dose in the amiodarone group. The other two groups were infused with the same volume of saline. After continued stimulus for 8 hours and maintained atrial fibrillation, the fight auricular tissue was taken for Western blot to examine the content of Cx40 and Cx43, the celluar electronic signal communication was detected by Lucifer Yellow scrape loading and dye transfer. Results In the rapid pacing group and amiodarone group, Cx40 and Cx43 levels decreased significantly ( P 〈 0.05 ) comparing with those in the normal control group. There was no significant difference between the amiodarone group and acute atrial fibrillation group for Cx40 and Cx43 levels ( P 〉 0. 05 ). Amiodarone can alter intracellular communication capacity between cardiac myocytes. Conclusions Cx40 and Cx43 have occured structure remodeling in dog with acute atrial fibrillation. Amiodarone can alter cell - to - cell communication, but it can not improve gap junction structure remodeling.
