强肝丸对肝纤维化大鼠肝组织胶原表达的影响

摘　　要：目的初步探讨强肝丸抗肝纤维化的药理作用。方法Wistar大鼠随机分成3组,即正常对照组、模型组、强肝丸治疗组。采用腹腔注射四氯化碳(CCl4)橄榄油混合液的方法制备肝纤维化动物模型。治疗组采用预防给药的方式,造模结束后持续治疗4周。治疗结束后,动物麻醉,取肝组织,进行羟脯氨酸含量检测、HE染色、Mallory染色及Ⅰ型胶原(CoL I)免疫组织化学染色。结果与模型组比较,强肝丸能显著降低肝组织中羟脯氨酸水平(P<0.01),减少肝组织中总胶原的合成和沉积(P<0.05),下调肝组织中Ⅰ型胶原(CoL I)的表达(P<0.05)。结论强肝丸对肝组织中胶原蛋白的合成过程有显著抑制作用,从而阻断肝纤维化的病理过程。Objective To investigate the pharmacological effects of Qianggan Pills in antagonizing hepatic fibrosis.Methods Wistar rats were randomly divided into 3 groups,i.e.the normal control group,model group and Qianggan Pills group.The model of hepatic fibrosis was established by intraperitoneal injection of mixture of CCl4 and olive oil.The Qianggan Pills group was treated for 4 weeks after the establishment of the model by using preventive therapy.When the treatment was finished the rats were anesthetized and their livers were taken out and stained with HE and Mallory stain methods.The content of hydroxyproline（HYP） was detected and collagen I was stained by immunohistochemical stain method.Results Comparing with the model group the level of HYP,synthesis and slugging of the total collagen were decreased significantly（P〈0.05）,and the expression of collagen I was down-regulated（P〈0.05） in Qianggan Pills group.Conclusion Qianggan Pills has a significant inhibitory effect on the synthesis of collagen protein in liver tissue,and it can interdict the pathological process of hepatic fibrosis.
