扶正祛瘀法对体外培养子宫肌瘤细胞凋亡调控基因表达的影响

摘　　要：目的探讨扶正祛瘀法治疗子宫肌瘤的分子学机制。方法体外原代培养子宫肌瘤细胞并传代后,加入扶正祛瘀中药,继续培养,以免疫细胞化学法检测细胞凋亡调控基因Bcl-2、Bax的表达。结果成功地进行了子宫肌瘤细胞的原代培养,加入扶正祛瘀中药后,Bcl-2表达明显下降(P<0.01),Bax表达明显升高(P<0.01)。结论扶正祛瘀法通过调控细胞凋亡调控基因表达,促进子宫肌瘤细胞凋亡,进而控制肌瘤的生长。Objective To discuss the molecule mechanism of the therapy of nourishing healthy qi and eliminating blood stasis in the treatment of uterine leiomyoma.Methods The primary cultured and passaged uterine leiomyoma cells were treated with the Chinese medicinals with the actions of nourishing healthy qi and eliminating blood stasis.The expressions of apoptosis regulating genes,i.e.Bcl-2 and Bax,were detected by using immunocytochemical assay.Results The cells from human uterine leiomyoma were primarily cultured successfully.After the administration of the Chinese medicinals with the actions of nourishing healthy qi and eliminating blood stasis the expression of Bcl-2 decreased significantly（P〈0.01）,and that of Bax increased significantly（P〈0.01）.Conclusion The therapy of nourishing healthy qi and eliminating blood stasis can improve the apoptosis of uterine leiomyoma cell,and inhibit the growth of uterine leiomyoma through regulating the expressions of apoptosis regulating genes.
