摘 要:目的研究倍他米松在促进早产胎鼠肺脏成熟的同时对早产胎鼠生长发育的影响及与IGF-Ⅰ的相关性。方法建立孕鼠模型,随机分为对照组,倍他米松单剂量组(SD-BTM),倍他米松多剂量组(MD-BTM),每组6只。在孕鼠怀孕14,15d给予不同剂量的倍他米松,在孕鼠怀孕16.5d行剖宫产终止妊娠,比较用药组与对照组胎鼠肺脏成熟和生长发育情况;放射免疫法测定孕鼠血清、胎鼠羊水中IGF-Ⅰ的含量,进行同组间的相关性分析。结果MD-BTM组出生体重、呼吸评分,SD-BTM组心脏质量/BW、全脑质量/BW高于对照组(P<0.05)。SD-BTM组、MD-BTM组胎鼠肺脏质量/BW、肝脏质量/BW,MD-BTM组全脑质量/BW均低于对照组(P<0.05)。SD-BTM组出生体重、呼吸评分,MD-BTM组心脏质量/BW与对照组比较无统计学意义(P>0.05)。SD-BTM组、MD-BTM组孕鼠血清IGF-Ⅰ含量低于对照组(P<0.01)。SD-BTM组胎鼠羊水IGF-Ⅰ低于对照组(P<0.01)。MD-BTM组胎鼠羊水IGF-Ⅰ与对照组比较无统计学意义(P>0.05)。孕鼠血清IGF-Ⅰ含量远高于胎鼠羊水IGF-Ⅰ含量,两者之间没有相关性。结论倍他米松在促进早产胎鼠肺脏成熟的同时影响孕鼠血清和胎鼠羊水IGF-Ⅰ含量。IGF-Ⅰ含量的变化影响胎鼠重要脏器的发育。Objective To investigate the effect of betamethasone on fetal rat growth and lung mature in the premature delivery rats and its correlation with insulin-like growth factor- Ⅰ ( IGF- Ⅰ ). Methods Pregnant rats were divided into three group: control group( n = 6), single dose of betamethasone group(SD-BTM group, n = 6), multiple doses of betamethasone group(MD-BTM group, n = 6). The pregnant rats were administrated with betamethasone at day 14 and 15,and then were performed by cesarean delivery at day 16.5. The lung mature and growth of fetal rats were compared. The IGF- I in serum of pregnant rats and fetal amniotic fluid were measured by radioimmunoassay. The correlation was analyzed. Results The birth weight and breath scale in MD-BTM group were higher than in control group(P〈0.05). The heart weight/birth weight and brain weight/birth weight ratios in SD-BTM group were higher than in control group ( P 〈 0.05). The lung weight/birth weight, liver weight/birth weight and brain weight/birth weight ratios were lower than in control group(P〈0.05). There was no significant difference in birth weight and breath scales between control group and SD-BTM group(P 〉0.05). There was also no significant difference in heart weight/birth weight between control group and MD-BTM group(P 〉 0.05). The serum IGF- Ⅰ of pregnant rats was lower than in control group(P 〈 0.05). The amniotic fluid IGF-Ⅰ of fetal rats was lower than in control group(P〈0.05). There was no significant difference in amniotic fluid IGF-Ⅰof fetal mice between control group and MD-BTM group(P 〉0.05). The serum IGF- I of pregnant rats was higher than that in amniotic fluid IGF-Ⅰ of fetal rats(P 〈 0.05), but there was no correlation between them. Condusion Antenatal treatment with betamethasone may improve lung growth and influence IGF- Ⅰ in serum of pregnant rats and amniotic fluid of fetal rats. The growth of major organs could be influenced through affecting IGF- Ⅰ
