用麻疹活疫苗制备抗麻疹病毒单克隆抗体及其应用

摘　　要：[目的]制备抗麻疹病毒(MV)的单克隆抗体(单抗,McAb),并用该单抗研制捕获法ELISA检测MV-IgM的诊断试剂盒。[方法]以MV减毒活疫苗免疫Balb/c小鼠,其脾细胞与SP2/0细胞融合,筛选分泌抗MV的杂交瘤细胞株,以获得该单抗,并对用其研制的检测抗MV-IgM试剂盒进行考核。[结果]获得的杂交瘤细胞株中,B2能稳定分泌高滴度抗MV单抗。经考核,用该单抗酶结合物研制的检测抗麻疹-IgM试剂盒特异性和稳定性良好,与同类商品试剂盒比较,其检出麻疹病例血清抗MV-IgM的时间较早。[结论]应用麻疹减毒活疫苗成功地制备了小鼠杂交瘤细胞,其中B2株能稳定分泌高效的抗MV单抗,用于研制检测抗MV-IgM捕获法ELISA试剂盒,其敏感性、特异性和稳定性令人满意。[Objective] To develop the monoclonal antibody （McAb} against measles virus（MV} and use it to prepare capture ELISA anti MV specific IgM kit. [ Methods] Balb/c mice were immunized with MV attenuated vaccines and their spleen cells were fused with SP2/0 cells. From these mice, the McAb against MV was obtained and used to prepare capture ELISA anti MV specific IgM kit. [ Results ] B2 strain hybridoma cells could secret McAb against MV in high titer specifically. McAb was used to preparate capture ELISA anti MV specific IgM kit. The specificity and stability of this kit were good. Anti MV - IgM in sera in cases with measles could be detected by this kit earlier than that by a same type commercial kit. [ Conclusion ] The sensitivity, specificity and stability of the capture ELISA anti MV specific IgM kit prepared using the McAb are satisfactory.
