摘  要:目的分析天津地区汉坦病毒分离株TJJ16 M基因的分子特征。方法采用细胞培养法从鼠肺标本中分离汉坦病毒TJJ16,提取TJJ16感染Vero-E6细胞的总RN A,经逆转录扩增病毒cDNA,继而进行PCR分别扩增M1和M2基因片段,克隆于T载体并测序拼接,对其进行序列分析和生物信息学分析。结果汉坦病毒TJJ16株的M基因片段全序列长为3616nt,仅有1个开放读码框架(ORF),编码1 136个氨基酸,序列分析表明其为汉滩型(HTN型)汉坦病毒,与HTN型Q32株同属1个亚型。结论TJJ16病毒株M基因片段的克隆和序列分析证实其为HTN型,系天津地区的首次报道。To explore the molecular characteristics of the M gene fragment from Hantavirus strain TJJ16 isolated in Tianjin area. This strain of virus was isolated by cell culture method, and the total cellular RNA was extracted from Veto-E6 cells infected with this strain. The viral M1 and M2 genes were amplified by RT-PCR, cloned into vector pMD18T and then underwent sequencing and splicing. The TJJ16 strain of M gene segment was found to be 3616 nt in length with one open reading frame(ORF) encoding 1136 amino acids, By the analysis of the M-segment sequence, it was demonstrated that this viral strain belonged to Hantavirus HTN type and was the same subtype as Q32 strain of HTN type. This strain of virus is the firstly reported strain of the HTN type of Hantavirus isolated in Tianjin area.
