高效液相色谱法检测乙酰基转移酶表型

摘  要:介绍了一种快速、简便的乙酰基转移酶表型的检测方法。口服100mg氨苯砜后3小时采血,用灵敏、特异的高效液相色谱法(分离柱:Spherisorb○RODS-C18)分别检测血浆中氨苯砜(DDS)及其乙酰化代谢产物单乙酰氨苯砜(MAD)的浓度,流动相为水∶乙腈∶冰乙酸=1060∶300∶25,紫外检测波长254nm时分离检测效果最好。DDS和MAD的平均回收率为84.5%和90.3%,最小检出浓度分别为0.001mg/L和0.01mg/L。An improved method for rapid and convenient determination of acetylator phenotype was described.The plasma concentrations of dapsone(DDS) and its acetylated metabolic product monoacetyldapsone(MAD) were measured three hours after a single dose oral intake of 100mg dapsone using the specific and sensitive high performance liquid chromatographic assay(Spherisorb ○R ODS C18).It was found that the separating efficiency was high when the mobile phase was composed of water∶acetonitrile∶acetic acid=1060∶300∶25.The detector's wavelength was adjusted to 254 nm and the flow rate was set at 1 0 ml/min.The average recovery ratios of DDS and MAD were 84 5% and 90 3%. The minimum detectable levels are as low as 0 001mg/L for DDS and 0 01mg/L for MAD,respectively.
