摘 要:[目的]主要研究不同提取条件对小油菜叶蛋白提取率的影响,确定小油菜叶蛋白提取的最佳工艺。[方法]以东北小油菜叶(干物)作为试验材料,分别利用凯式定氮法、索式抽提法和直接干燥法测定油菜叶中叶蛋白、粗脂肪和水分含量,研究不同pH值、提取温度、时间及料液比对叶蛋白提取率的影响。[结果]在pH值9,温度45℃,固液比1∶30,浸提时间1 h时,蛋白质提取率达88.20%。[结论]该研究确定了小油菜叶蛋白提取的最佳工艺,为当地小油菜天然叶蛋白生产的开发提供了科学依据。[Objective]Different extraction conditions which influenced extraction percentage of Northeast Rape leaves was studied to ascertain the best extraction technics of protein in Northeast Rape leaves.[Method] Northeast Rape leaves(dry) as subject,using the Kjeldahl Nitrogen Determination method,Soxhlet extraction method and direct drying method,the content of leaf protein,crude fat and water in Northeast Rape leaves were determined to study the influence of different pH,extraction temperature,time and the ratio of solid-liquor on the extraction percentage of leaf protein.[Result] When pH value was 9,temperature was 45 ℃,ratio of solid-liquid was 1∶30,and lixiviation time was 1 hour,the extraction percentage of protein reached 88.20%.[Conclusion] The best technics of leaves protein of Northeast Rape was determined.Scientific basis was provided to explore local rape natural leaf protein production.
