摘 要:目的探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)对豚鼠透镜诱导近视眼(lens inducedmyopia,LIM)后极部巩膜细胞凋亡的影响及其在抑制近视形成中的作用机制。方法将4周龄的清洁级健康花色豚鼠随机分为正常对照组(双眼均不处理)、LIM 组、LIM+PBS 组、LIM+bFGF 低剂量(100 ng)组、LIM+bFGF 中剂量(500 ng)组和LIM+bFGF 高剂量(1000 ng)组,每组15只。除正常对照组外,其余各组右眼(实验眼)配戴-10 D 透镜7 d 后,将相应浓度的 bFGF 或 PBS 注入豚鼠玻璃体腔,3 d 后再次注射,配戴透镜15 d 后,取后极部巩膜组织,用电镜、终末脱氧核糖核苷酸转移酶介导原位缺口末端标记法(TUNEL)及流式细胞术检测巩膜的凋亡细胞,应用 Ki-67免疫组织化学染色检测巩膜细胞增殖的情况。结果与正常对照组比较,LIM 组处理眼有明显近视形成[(3.57±0.78)D]和巩膜细胞的凋亡[凋亡率为(11.28±0.04)%],差异有统计学意义(P<0.05);与 LIM+PBS 组比较,LIM+bFGF 低剂量组、LIM+bFGF 中剂量组和 LIM+bFGF 高剂量组处理眼近视被抑制,凋亡率明显降低,差异均有统计学意义(P<0.05)。随着 bFGF 剂量加大,bFGF 抑制近视的作用增强。结论 bFGF 能通过降低后极部巩膜细胞凋亡率有效抑制豚鼠 LIM 的形成。Objective To investigate the effect of basic fibroblast growth factor (bFGF)on the apoptosis of scleral cells in the posterior pole in lens-induced myopia of guinea pigs and to discuss the mechanism of bFGF in inhibiting the formation of myopia. Methods Four-week-old cleaning healthy guinea pigs were randomly divided into the control group, LIM group, LIM+ PBS group, LIM+ the bFGF 100 ng group,LIM+ the bFGF 500 ng group, LIM+ the bFGF 1 000 ng group,15 in each. Except the control group, the right eye of guinea pig in other groups wore -10 D concave lens for 7 days and then different concentration of bFGF or PBS were injected into the vitreous cavity,3 days later injected again. After 15 days of -10 D concave lens treatment,the eyeballs were removed and the apoptotic cells in the scleras were determined by electron microscopy ,TUNEL technique and flow cytometry. The proliferation of scleral cells in the posterior pole were determined by Ki-67 immune histochemistry stain. Results Compared with the control group, the significant differences were detected in the right eyes of LIM group (P〈0.05) ,which showed myopia [(3.57± 0.78)D] and the apoptosis of scleral cells [the apoptotic rate was (11.28±0.04)%].Compared with LIM+PBS group ,eye myopia was inhabited and the apoptotic rate of scleral cells decreased in LIM+ the bFGF 100 ng group, LIM+ the bFGF 500 ng group, LIM+ the bFGF 1 000 ng group,which showed significant differences (P〈0.05).With the increasing of bFGF,the inhibiting action increased. Conclusion Infusing bFGF into the vitreous cavity may decrease the apoptotic rate of scleral cells in the posterior pole and inhibit the formation of LIM effectively.
