摘 要:目的探讨葡多酚(GPC)和硫辛酸(LA)对^(60)Coγ射线照射小鼠肝细胞导致自由基损伤的拮抗作用。方法将10只1周龄清洁级昆明种小鼠肝脏制成1×10~9个几肝细胞培养物,细胞存活率均>85%。将肝细胞培养液分为阴性对照组(未经^(60)Coγ线照射)、GPC 组(终浓度为75 mg/L)、LA 组(终浓度为100 mg/L)和阳性对照组(不含任何抗氧化剂),GPC组、LA 组和阳性对照组给予剂量为2.5 Gy 的^(60)Coγ射线照射,距离90 cm,持续10 s。检测肝细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力和丙二醛(MDA)含量,并进行彗星试验。结果与阳性对照组比较,GPC 组和LA 组肝细胞 SOD、GSH-Px 活力均较高(P<0.05或 P<0.01),MDA 含量均较低(P<0.01),彗星面积较小(P<0.05),尾长较短(P<0.01),拖尾率较低(P<0.05和 P<0.01)。结论葡多酚和硫辛酸对^(60)Coγ射线照射小鼠肝细胞导致的自由基损伤有拮抗作用。Objective To study the antagonistic effects of grape procyanidin (GPC) or lipoic acid (LA) to the rat liver cell peroxidative damage induced by^60Coγ radiation. Methods The cultured rat liver cells( 1×10^9/L )were prepared from 10 rats aged 1 week, the survival rate was above 85%. The cultured liver cells were divided into the negative control group (without radiation), GPC group (75 mg/L) ,LA group ( 100 mg/L)and the positive control group (containing no antioxidants ),which were radiated by ^60Co γ radiation(2.5 Gy,90 cm,10 s).The SOD,GSH-Px activity, MDA level and comet assay were determined. Results Compared with the positive control group,in the GPC group and LA group, the activity of hepatic SOD and GSH-Px were significantly higher (P〈0.05 and P〈0.01 ) respectively, the MDA level was significantly lower (P〈0.01),the comet field and tail length were significantly lower (P〈0.05 and P〈0.01 ) respectively. Conclusion Grape procyanidin or lipoic acid has a significant antagonistic effect against the liver cell peroxidative damage induced by ^60Coγ radiation.
