鸭源巴氏杆菌-大肠埃希菌二联多价蜂胶灭活疫苗的制备与免疫效果试验

摘　　要：本研究选取鸭致病性大肠埃希菌(E.coli)常见血清型O1、O2、O18、O78和3株分离自发病鸭场、毒力强、免疫原性好的巴氏杆菌(Pasteurella multocida,Pm)D22、D23、D157作为制苗菌株,分别以鲜血琼脂和马丁琼脂进行固体表面培养制备的抗原与蜂胶佐剂高速乳化,制备鸭多杀性巴氏杆菌-大肠埃希菌二联多价蜂胶灭活疫苗。结果表明,疫苗安全可靠,雏鸭一免后第14天对同源E.coli、Pm攻击免疫保护率为100%和90%;雏鸭二次免疫后第14天的免疫保护率均为100%。用间接ELISA对免疫鸭进行抗体消长规律检测,7日龄商品麻鸭首免后20 d抗体达到高峰,抗体维持时间在54 d以上;首免7 d后进行二免,可产生显著高于一免的抗体水平。疫苗免疫雏鸭能够显著(P<0.05)或极显著(P<0.01)地增强T细胞的免疫力,可维持3周左右。疫苗在4℃可保存12个月以上。This research used duck pathogenic Escherichia. coli serotyoes O1,02,018,078 and 3 strains of Pasteurella multocida D22,D23,D157 which were isolated from dead or diseased ducks. These strains had strong pathogenicity and good immunogenicity. We used them as vaccine-making bacteria, to prepare antigens by solid surface culture on blood agar medium and Martin agar medium,and then added propolis adjuvant,emulsified with high speed so as to obtaine combined polyvalent propolis inactived vaccin. The result demonstrated that the vaccine is safe and reliable, immunized ducks which were challeged by homologous of P. m and E. coli demonstrated that 14 days after vaccination, the immunoprotective rates were 90% and 100%,14 days after revaccination, ducks had strong immunity, the immunoprotective rates were both 100%. Indirect enzyme-linked immunosorbent assay （ELISA） showed that specific antibody peaked 20 days post vaccination in titer and could sustain over 54 days post vaccination. Antibody level in the revaccination group was higher. The notably enhanced cell mediated immunity in the experiment group was significantly different （P 〈0.05 or P 〈 0.01） from the control group and was about 3 weeks in duration. The vaccine can be stored in 4 ℃ over 12 months.
