摘 要:H2O2能够氧化茜素蓝S使之褪色,而模拟酶——血红蛋白对其具有催化作用。据此建立了一种以茜素蓝S为指示剂的过氧化氢——茜素蓝S——血红蛋白酶催化反应体系测定痕量H2O2的新方法。研究了反应的最佳条件,体系的酸度为pH=10.1的NH3-NH1Cl缓冲溶液,最大吸收波长为580nm。该法的线性范围为2.0×10^-7.5×10^-5mol/L,检出限为1.3×10mol/L,表观摩尔吸光系数为1.7×10^1L/(mool·cm)。该方法可用于雨水及消毒水中H2O2的测定。结果今人满意。该方法简单、灵敏、选择性好,易于操作。A new enzymatic catalytic method was developed for the determination of trace hydrogen peroxide using hemoglobin as catalyst. The color of alizarin blue S was faded by oxidation with hydrogen peroxide. The oxidation was enhanced in the presence of hemoglobin. The optimum conditions and effect of foreign ions were discussed. At pH 10.1 of NH3-NH4CI buffer solution, the system exhibited maximum absorption wavelength at 580 nm. Under the optimal conditions, the degree of fading (AA) was linear to the H2O2 concentration (c) in the range of 2. 0 × 10^-7 - 7. 5 × 10^-5 mol/L, and the apparent molar absorptivity εszs was 1.7× 10^4 L/(mol · cm) with the detection limit of 1.3 × 10^-8 mol/L. This method is simple, sensitive and suitable for the determination of hydrogen peroxide in rain and disinfector with satisfactory results.
