摘 要:本文研究了钙结合蛋白S100A8在辐射损伤小鼠骨髓细胞中的表达,及其与细胞周期、凋亡的关系。运用蛋白/DNA双参数流式细胞技术,比较3.5、7Gy60Coγ射线照射后6h小鼠骨髓中S100A8蛋白的表达变化,及S100A8阳性(S100A8+)与S100A8阴性(S100A8-)细胞周期和凋亡的改变。正常小鼠骨髓中,S100A8+细胞主要是粒细胞;与S100A8-细胞相比,S100A8+细胞增殖不活跃。辐射损伤后早期,小鼠骨髓中S100A8+细胞数及S100A8蛋白表达量无明显改变,S100A8-细胞发生G2/M期阻滞及凋亡,而S100A8+细胞周期不改变,也不发生凋亡。结果表明,S100A8+细胞相对S100A8-细胞具有辐射抗性。The study explores the expression of calcium-binding protein S100A8 and the relationship between S100A8 and cell cycle or apoptosis in bone marrow cells of mace with radiation injury. The protein/DNA bi-pararneter flow cytometry method has been used to analyze the change of S100A8 protein expression and the difference in cell cycle or apoptosis between S 100A8 positive and S 100A8 negative cells in bone marrow of mace at 6h after irradiation with 3.5 or 7 Gy ^60Co γ-ray in vivo. In the normal bone marrow of mice, S100A8 positive ceils are mainly granulocytes, and they are inactive in cell proliferation compared to S100A8 negative ceils. At the early pfiase of radiation injury, the proportion of S 100A8 positive cells and the expression level of S 100A8 protein have no sig- nificant change in bone marrow of mace; G2/M arrest and apoptosis occur in S 100A8 negative ceils, but no change of cell cycle and apoptosis occur in S 100A8 positive ceils. These phenomena suggest that S 100A8 positive cells in bone marrow of mice are radiation resistant compared to S 100A8 negative cells.
