摘 要:目的观察连黛片对实验性HT29大肠癌裸鼠移植瘤端粒酶活性及其相关基因hTERTmRNA表达的影响。方法采用皮下注射HT29大肠癌细胞株制作结肠癌裸鼠模型,每组均在初次接种后7天进行灌胃治疗,连续灌胃1个月。中药治疗组用连黛片灌胃治疗(每只灌胃2g/kg),西药治疗组用端粒酶抑制剂3’-叠氮3’-脱氧胸腺核苷(叠氮胸苷,AZT,150mg/kg)灌胃治疗,空白对照组予以生理盐水灌胃。动态观察各组裸鼠肿瘤体积变化,用游标卡尺测量每组裸鼠移植瘤的大小。采用TRAP-ELISA方法检测端粒酶活性变化,荧光定量PCR检测端粒酶hTERTmRNA的表达,结果进行统计学分析。结果①初次接种后7天各组裸鼠皮下肿瘤大小比较无显著性差异;②1个月后治疗组裸鼠皮下肿瘤体积比较无明显差异,治疗组与空白对照组比较有差异;③空白对照组端粒酶相对活性较两组治疗组明显增强;空白对照组端粒酶hTERTmRNA的表达增多。中药组与AZT组端粒酶活性及端粒酶hTERTmRNA的表达改变无显著性差异。结论连黛片能抑制人大肠癌HT29裸小鼠移植瘤的增值,其作用机制可能是减少肿瘤细胞hTERTmRNA的表达,从而降低肿瘤组织端粒酶活性。Objective To observe the influence of "Liandai Tablet" on the telomerase activity and the expression of hTERT mRNA in local tumor tissues of experimental nude mice. Methods The model of colonic cancer of nude mice were replicated by injecting HT29 colonic cancer cell subcutaneously. Every nude mouse was lavaged after the first injection for one month. Chinese herb group was lavaged with "Liandai Tablet"(2g/kg), western drug group with AZT (150 mg/kg), blank control group with 0.09% sodium chloride. The change of tumor volume of nude mice was observed; the telomerase activity was detected with TRAP-ELISA, and the expression of hTERT mRNA with fluorescent quantitative PCR. Results There was no significant difference between the three groups in tumor volume of nude mice seven days after the first inoculation; there was no significant difference between treatment groups about tumor volume of nude mice subcutaneously after one month, but there was a difference between treatment groups and control group; there was no significant difference between treatment groups in the change of telomerase activity and the expression of hTERT mRNA, but there was a significant difference between treatment groups and control group. Conclusion "Liandai Tablet" can restrain the proliferation of transplantation tumor of nude mice of HT29 cell strain of colon tumor possibly by reducing the expression of hTERT mRNA and lowering the telomerase activity in local tumor tissues.
