摘 要:为研究地高辛标记和生物素标记核酸探针斑点杂交检测HBVDNA的敏感性、特异性和稳定性,用两种探针和(32)P标记探针进行平行检测对照.结果表明,地高辛探针检测灵敏度为0.1Pg,已达到(32)P中标记探针水平。生物素探针为1~10Pg。两种探针与32P标记探针相比。符合率分别96.65%和89.13%;敏感性分别为94.44%和83.33%;特异性分别为96.43%和92.86%。两种探针在-20℃存放6月后检测结果前后一致。研究结果表明,地高辛标记探针和生物素标记探针斑点杂交检测HBV.DNA都具有较高的敏感性、特异性和稳定性,其中以地高辛标记探针更优。To investigate the sensitivity, specificity and stability of the Dig-labelled and Biotin-labelled nucleiotide probes in the dot hybridization detection of HBV DNA. thses two probes were used to detect HBV DNA separately, and a 32 P-labelled probe was used as the control. The sensitivity of the Dig-labelled probe was 0. 1Pg, which was as high as that of the32 P-labelled, labelld was 1-10Pg. Compared with the 32 P-labelled probe, the positive coincidence cates of the Dig-labelled probe and the Biotin-labelled probe were 96. 65% and 89.13%, respectively; the sensitivities of them were 94. 44% and 83. 33%, respectively;and the specificities of them were 96.43% and 92. 86%, respetiveely. The detection results of thses two probes before before and after their storage at-20℃ for -6 months. had on difference. Both the Dig-labelled and Biotinlabelled probes had high sensitivities,specifitciyies and stabilities in detecting HBV DNA, but the Dig-labelled probe was better. biotin-labelled probe.
