摘  要:目的研究氯乙烯(VC)对小鼠外周淋巴细胞DNA的损伤作用及对肝脏功能的影响,探讨VC所致损伤的早期敏感监测指标。方法32只雄性小鼠随机分成4组,染毒剂量分别为0,10,20,40 mg/kg,连续腹腔注射染毒4周,隔2天染毒一次。DNA损伤采用单细胞凝胶电泳实验(彗星实验)法进行评价,并测定丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)以反映肝功能变化。结果染毒组小鼠外周淋巴细胞的彗星率均高于对照组(χ2=251.17,P<0.01),彗星细胞尾相值(oliver tail moment,OTM值)亦显著高于对照组(χ2=557.760,P<0.01),各染毒组与阴性对照的肝功能差异均无统计学意义。结论本实验条件下,在未导致肝功能变化时,VC可导致外周淋巴细胞DNA损伤。彗星细胞的损伤程度及出现率可作为氯乙烯所致外周淋巴细胞DNA损伤的早期监测指标。Objective To investigate the effects of vinyl chloride (VC) on liver function tests and lymphocyte DNA in mice. Methods 32 healthy male mice were randomly divided into four groups: 10, 20, 40 mg/kg and control group. The mice were treated with gaseous VC by intraperitoneal injection with two day interval for 4 weeks. Then, single cell gel electrophoresis ( SCGE), ALT, AST and ALP examinations were applied for detecting DNA damage of peripheral lymphocytes and hepatic function. Results The comet cells rates in experimental groups were significantly higher than that of control group (χ^2 = 251.17, P 〈 0. 01 ), and the difference in average OTM ( Oliver tail moment) of comet in the exposed groups was significant (χ^2 =557. 760, P 〈0. 01 ). However, there was no significant difference in changes of liver function between exposed groups and control group. Conclusions VC can cause DNA damage of lymphocytes, which might be an early monitoring index of the DNA damage induced by VC.
