苯甲酸雌二醇对小鼠生殖器官肥大细胞的影响

摘　　要：为研究苯甲酸雌二醇(E2)对小鼠生殖器官肥大细胞数量和分布的影响,将4周龄小鼠随机分成2个对照组和3个试验组,每组10只(雌雄各半),连续腹腔注射E2(0.05、0.5、5mg.kg-1.d-1)7d后手术采取各组睾丸、附睾、卵巢和子宫,组织经PBS配置4%多聚甲醛固定24h,石蜡包埋,5μm厚切片,采用改良甲苯胺蓝染色法(MTB)观察生殖器官中肥大细胞数量分布。结果表明,注射0.05mg.kg-1.d-1剂量使子宫肥大细胞增多,注射0.5mg.kg-1.d-1和5mg.kg-1.d-1剂量均使子宫肥大细胞减少;注射这3种剂量均使卵巢肥大细胞增多;注射0.05mg.kg-1.d-1和0.5mg.kg-1.d-1剂量使睾丸和附睾肥大细胞增多,注射5mg.kg-1.d-1剂量却使睾丸和附睾肥大细胞减少。To investigate the effects of estradiol benzoate （E2） on the number and distribution of mast cells in mice reproductive organs, fifty mice （four-week old） were randomly divided into five groups, namely two control groups and three treatment groups, with 10 mice （half of each sex） in each group. Mice had been administered （0.05, 0. 5, 5mg · kg^-1· d^-1 with E2 by celiac injection for 7 days. The samples of testis, epididymis, ovary, and the uterus were then collected by surgical operation. The tissues samples were fixed in 4% paraformaldehyde in phosphate-buffered saline for 24h and then processed to paraffin wax. Five - micron - thick sections were cut for mast cell detection. The distribution and number of mast cells in the reproductive organ of mice were determine by using modified toluidine blue （MTB） staining. The results showed as follows：0.05mg · kg^-1· d^-1 dose of E2 administrated significantly increase uterus mast cells, but 0.5mg · kg^-1· d^-1 and 5mg · kg^-1· d^-1 dose of E2 significantly decreased uterus mast cell. All doses of E2 administrated significantly increased ovary mast cells. 0.05mg · kg^-1· d^-1 and 0.5mg · kg^-1· d^-1 dose of E2 administrated increased the mast cell number in the testes and epididymis,but 5mg · kg^-1· d^-1 dose of E2 significantly decreased the mast cells in testicle.
