摘  要:目的:观察补肾阳中药复方对小鼠胚胎干细胞活动的影响。方法:选用小鼠胚胎干细胞模型CRL-1825细胞株,加入补肾阳中药复方大鼠含药血清,观察补肾阳中药对干细胞增殖、分化、凋亡以及相关基因表达的影响。结果:补肾阳中药可以抑制CRL-1825细胞凋亡,但对CRL-1825细胞增殖、分化无明显影响;补肾阳中药同时可促进CRL-1825细胞Notch1基因表达,相对表强度由46.33升高至102.81;补肾阳中药还可以抑制P16INK4a基因表达,相对表强度由202.11降至93.11。结论:补肾阳中药复方可以抑制CRL-1825细胞凋亡,并可能与调控Notch1、P16INK4a表达相关。Objective: To observe effects of nourishing kidney-yang on activities of embryonic stem cell. Method: Embryonic stem cell line CRL-1825 was used as a model. Serum of kidney-yang-nourishing herbals was added to CRL-1825 medium, and the effects of kidney-yang-nourishing herbals on CRL-1825 cell proliferation, differentiation, apoptosis and related gene expression were observed. Result: Kidney-yang-nourishing herbals could inhibit the CRL-1825 cell apoptosis, without influences on CRL-1825 cell proliferation and differentiation; it also up-regulates Notchl expression and down-regulates PI6INK4a expression. Conclusion: Nourishing kidney-yang could inhibit CRL-1825 cell apoptosis, it may relate to Notchl and P16INK4a expression.
