摘  要:目的研究OD-PTD融合蛋白对K562细胞的作用。方法应用细胞计数的方法研究OD-PTD融合蛋白对K562细胞的生长曲线的影响;用MTT研究OD-PTD融合蛋白对K562细胞的细胞毒作用;用流式细胞仪技术研究OD-PTD融合蛋白对K562细胞周期的影响。结果OD-PTD融合蛋白能显著抑制细胞的生长(P<0.01);并且对K562细胞具有明显的细胞毒作用(P<0.01)。OD-PTD融合蛋白作用于K562细胞后,细胞周期变化明显,细胞从G0/G1期到S期发生明显阻滞(P<0.05)。结论OD-PTD融合蛋白对K562细胞有明显的抑制作用,为进一步探讨慢性粒细胞白血病新的治疗手段提供了实验依据。Objective To explore the effects of OD-PTD fusion protein on K562 cells.Methods Proliferation ability of K562 cells treated by OD-PTD fusion protein was studied by growth curve. The cytotoxicity of OD-PTD fusion protein to K562 cells was determined by MTT assay. K562 cells' cycle change induced by OD-PTD fusion protein was investigated by flow cytometry. Results OD-PTD fusion protein significantly inhibited the growth of K562 cells, and it had a remarkablely cytotoxic effect on K562 cells. After several hours of exposure to OD-PTD fusion protein, the progression of cell cycle was arrested from G0/G1 to S phase. Conclusion OD-PTD fusion protein can significantly inhibit the proliferation of K562 cells, which provides an experimental basis for new therapy of chronic myeloid leukemia.
