摘  要:采用液体摇瓶培养方法,探讨了金属离子、pH值、培养温度、碳源、氮源、表面活性剂、诱导剂等各种因素对白腐菌(Pleurotus ostreatus)分泌漆酶能力的影响,采用正交实验对主要的影响因素进行了优化。结果表明,Cu2+、Mn2+的加入能促进白腐菌分泌漆酶,Ag+、Fe2+严重抑制白腐菌分泌漆酶,最佳碳源为葡萄糖、最佳氮源为蛋白胨,表面活性剂Tween-80和诱导剂的加入能显著提高漆酶的分泌。优化的培养条件为:土豆汁20%,葡萄糖20 g.L-1,蛋白胨4 g.L-1,CuSO40.5 mmol.L-1,KH2PO43 g.L-1,MgSO41.5 g.L-1,ZnSO450 mg.L-1,MnSO450 mg.L-1,维生素B12.0 mg.L-1,Tween-80 0.5 g.L-1,ABTS或XYL 1 mmol.L-1,pH值为4,28℃下摇瓶培养9 d,此时,漆酶酶活可达21 300 U.L-1。The main factors affecting laccase production by Pleurotus ostreatus in liquid shake-flask fermentation were studied. The effects of metal ion, pH value culture, culture temperature, carbon source, nitrogen source,surfactant, inducer on producing laccase of Pleurotus ostreatus were studied. The optimization of fer-mentation conditions were conducted according to the orthogonal experiment. The results showed that,addition of Cu^2+ and Mn^2+ could improve the production of laccase, but Ag^+ and Fe^2+ inhibited it remarkably. The optimum carbon source and nitrogen source were glucose and peptone,respectively. And the capabilities of producing laccase were evidently enhanced with the addition of Tween-80 or inducer such as ABTS and XYL. The optimal fermentation condition for laccase production were as follows:potato juice 20% ,glucose 20 g·L^-1 , peptone 4 g·L^-1 , CuSO40.5 mmol·L^-1 , KHzPO43g·L^-1 , MgSO41.5 g·L^-1 , ZnSO4 50 mg·L^-1 , MnSO4 50 mg·L^-1 , vitamin B1 2.0 mg· L^-1 ,Tween-80 0.5 g· L^-1 ,ANTS or XYL 1 mmol·L^-1 , pH value of 4, and incubation with 130 r ·min^-1 at 28℃ for 9 days. Laccase activity was 21 300 U ·L^-1 under the optimal fermentation conditions.
