Ad5F35-LMP2重组腺病毒修饰DC体外诱导特异性T细胞免疫的研究

摘　　要：目的了解含有EB病毒潜伏膜蛋白2的非复制型重组腺病毒（Ad5F35-LMP2）修饰的DC能否在体外诱导LMP2特异性细胞毒性T淋巴细胞免疫。方法人外周血单个核细胞在细胞因子诱导下生成树突状细胞，Ad5F35-LMP2感染树突状细胞后激发同源的T细胞，MTT法检测其对T细胞的增殖作用，及激活的特异性CTL对表达LMP2的人CNE-2细胞的特异性杀伤效应。结果Ad5F35-LMP2能有效感染树突状细胞，其激活的特异性CTL对CNE-2细胞有特异性杀伤活性，与对照组相比差异有统计学意义（P〈0．01）。结论重组腺病毒Ad5F35-LMP2感染的DC可以有效的诱导产生EBV-LMP2特异性细胞毒效应。Objective To observe whether dendritic cells（DCs） transfected with recombinant adenovirus AdSF35-LMP2 induces LMP2 specific immunity mediated by cytotoxic T lymphocytes in vitro. Methods Dendritic cells have been generated in vitro, and cocuhured with autologous T cell after the DCs were infected with AdSF35-LMP2, then the proliferation of the induced T cells and their cytotoxic activity against CNE-2 tumor cells which express EBV-LMP2 protein on membrane were assessed by MTT ethod. Results The dendritic cells could be transfected with AdSF35-LMP2 and the CTL activated by AdSF35-LMP2-DC could effectively suppress the proliferation of CNE-2 cells compared with control groups. Conclusion The dendritic cells transfected with recombinant adenovirus AdSF35-LMP2 showed cytotoxicity effect by activating T lymphocytes.
