食管癌中Survivin超微结构定位及其在组织中的表达

摘　　要：目的:研究抗凋亡基因Survivin在食管癌细胞中超微结构的定位及表达情况,以探讨Survivin基因在食管癌发病机制中的作用。方法:应用免疫电镜胶体金标记对3例新鲜食管癌组织Survivin蛋白进行超微结构定位观察,采用免疫组化技术检测30例食管癌及其对应的癌旁正常组织中Survivin蛋白的表达。结果:透射电镜下观察食管癌细胞结构清晰,金颗粒呈团状、点灶状分布,Survivin蛋白可定位于线粒体、内质网及胞浆中,也可定位于胞核、核仁、核膜及核周间隙中;免疫组化结果示,食管癌组织中Sur-vivin阳性表达率为80.00%,明显高于癌旁组织6.67%(P<0.01),并与食管癌的分化程度、临床分期、淋巴结转移有关(P<0.05,P<0.01)。结论:免疫胶体金标记Survivin基因在食管癌中可准确定位,Survivin蛋白异常高表达与食管癌的发生、发展密切相关,对食管癌的早期诊断是一个重要的参考指标。Objective：To detect the expression of Survivin in esophageal cancer and ultrastrutural lacalization with immunoelectron microscope and to investigate its molecular mechanism involved. Methods： Colloid gold labeling and immunoelectron microscope technique were used to locate and observe ultrastructure of Survivin in esophageal cancer. Expression of Survivin was detected in paired normal and tumor tissues from patients with esophageal cancer by immunohistochemistry. Results：Esophageal mucosa was clear under transmission electron microscope. Survivin colloid gold particles were distributed in spotty group in esophageal cancer. It was located in endoplasmic reticulum, chondriosome, kytoplasm, nucleolus karyotheca, karyon. The positive rate of surviving expression in esophageal carcinoma and normal mucosa adjancent to tumor was 80.00% and 6.67% , respectively, which was obviously higher in the former than in the latter（ P 〈 0.01 ）. There was a significant different in the positive rates of Survivin expression between the low and high differentiated esophageal carcinomas （ P 〈 0. 05, P 〈 0.01 ）. Conclusion：Survivin can be precisely located with Colloid gold labeling esophageal cancer cells surviving. The expression of Survivin is significantly related to the genesis and progression of esophageal carcinoma, which is an important reference index of early diagnosis.
