摘  要:通用Nafion固定二茂铁(Fc)作为电子传递体,通过明胶包埋法将葡萄糖氧化酶(GOD)和α-糖苷酶(GA)修饰在Nafion-二茂铁修饰的玻碳电极上,制成检测脑疲劳标志物—唾液淀粉酶的生物传感器。结果表明:该传感器表现出良好的特异性、敏感性、准确性,传感器响应电流与淀粉酶浓度在50~820U/L之间呈现良好的线性关系,检出限为17.5U/L,达到95%稳态响应时间不超过30s。探讨了pH、温度及其他干扰物质等对淀粉酶生物传感器的影响。将此传感器用于唾液中淀粉酶的测定,取得了满意的结果。Biosensor is developed for detecting the marker of brain fag-amylase. Ferrocene, as the electron transfer,is immobilized by Nation. Glucose oxidase (GOD)and α-glucosidase (GA)are linkes on the Nation- ferrocene modified glassy carbon electrode by gelatin embedment. The results show that the biosensor displays a good specificity, sensibility and accuracy. The current of the biosensor has a linear relationship with the concentration of amylase about 50 -820 U/L. The detection limit is 17.5 U/L and response time achieved 95 % steady state is no more than 30 s. The biosensor response is investigated in terms of pH ,temperature and etc. The biosensor is applied for direct measurements of amylase in saliva of human, and satisfactory result is got.
