摘 要:目的:研究蛋白酶体抑制剂MG132对沙眼衣原体感染细胞促凋亡蛋白Bim、Puma表达的影响,探讨沙眼衣原体抑制宿主细胞凋亡的可能机制。方法:以沙眼衣原体(D血清型)感染Hela229细胞,用不同浓度MG132作用后,Western-blot检测Bim、Puma蛋白质表达水平的变化。结果:沙眼衣原体感染12h后Bim、Puma的表达降低;24h后完全消失。MG132能抑制Bim、Puma的表达下降。结论:沙眼衣原体感染Hela229细胞后可降低促凋亡蛋白Bim、Puma蛋白质的表达;MG132可阻止这一作用,Bim和Puma的降解依赖蛋白酶体的活性。Objective:To explore the effects of MG132 on Proapoptotic Proteins Bim,Puma and the mechanism of apoptotic inhibition in Chlamydia trachomatis-infected Hela229 cells. Methods:The experiments cell were treated with the proteasome inhibitor MG132. Western-blot was applied to detect the expression of Bim,Puma at protein level. Results:Bim, Puma were degraded approximately 12 h after infection with C. trachomatis and disappeared at 24 h. MG132 can inhibit the degradation. Conclusion:The levels of Bim,Puma were reduced by infection with C. trachomotis; the reduce was prevented by MG132. Degradation of Bim,Puma requires proteasomal activity.
