摘 要:目的:探讨visfatin对血浆胆固醇代谢和肝胆固醇合成的影响.方法:将空载体转染对照组(NC组),大鼠visfa-tin基因真核表达质粒转染普通饲料组(NT组)和高脂组(HT)大鼠,并测定转染前后各组大鼠血浆胆固醇及肝脏HMG CoA还原酶和固醇调节元件结合蛋白2(SREBP2)mR-NA表达的变化.结果:visfatin质粒注射3 d后,NT和HT组大鼠血浆胆固醇均较注射前明显下降[(1.61±0.18)vs(1.25±0.09)mmol/L和(2.37±0.11)vs(1.74±0.12)mmol/L,均P<0.05];高密度脂蛋白胆固醇(HDL-C)也明显降低[(0.83±0.09)vs(0.56±0.09)mmol/L和(1.32±0.09)vs(0.91±0.04)mmol/L,P<0.05];NT和HT组大鼠SREBP2mRNA表达均明显高于NC组[(0.37±0.06),(0.36±0.07)vs(0.21±0.04),均P<0.05],NT组HMG CoA还原酶mRNA水平明显高于NC,HT组[(0.89±0.13)vs(0.37±0.07)和(0.55±0.16),均P<0.05],NC组与HT组无明显差异.结论:visfatin可能通过促进SREBP2和HMG CoA还原酶表达来调节体内胆固醇代谢.AIM: To investigate the effects of visfatin on hepatic cholesterol metabolism and the expression of SREBP-2 gene in rat. METHODS: The control plasmid pcDNA3.1 ( ± ) and recombinant plasmid pcDNA3. 1-visfatin were transfected into rats in control group(NC) ,normal diet group(NT) and high-fatty-diet group( HIT), respectively. The plasma cholesterol, hepatic HMG CoA reductase and sterol regulatory element binding protein 2 (SREBP2) mRNA were evaluated by RT-PCR before and after transfection. RESULTS: Plasma total cholesterol (TC) [ ( 1.61 ± 0. 18)vs(1.25 ±0.09 )mmol/L and (2. 37 ±0. 11)vs ( 1.74 ± 0. 12) mmol/L,P 〈0.05 ] and high-density lipoprotein cholesterol(HDL-C) [ (0.83 ± 0.09) vs (0.56 ± 0. 09) mmol/L and (1.32±0.09) vs (0.91 ±0.04) mmol/L, P〈0.05]Of group NT and HT were significantly decreased after 3 d of pcDNA3.1- visfatin transfection; SREBP2 mRNA [ ( 0. 37 ± 0. 06 ) and (0.36 ±0.07) vs(0. 21±0.04) ,P 〈0.05] and HMG CoA reductase mRNA[ (0.89 ± 0.13 ) and (0.55 ± 0.16) vs (0.37 ± 0.07 ), P 〈0.05 ] levels were significantly higher than those of group NC, and HMG CoA reductase mRNA level of group NT was higher than that of group HT, but there was no significant differ-ence between group NT and HT in SREBP2 mRNA levels. CONCLUSION: Visfatin regulates cholesterol metabolism by enhancing expression of SREBP2 and HMG COA reductase.
