摘 要:目的:通过观察氟喹诺酮类药物和糖皮质激素类药物作用于大鼠跟腱后的细胞学表现,验证两药是否对跟腱有损伤作用,以及两者是否具有协同效应。方法:实验于2007—08/09在甘肃中医学院实验动物中心完成。①实验材料:SPF级健康成年Wistar大鼠40只,雌雄各半,体质量200~250g。实验用药盐酸环丙沙星片(国药准字H23022275)为哈药集团生产;醋酸泼尼松龙注射液(浙卫药准字(1996)第185301号)为浙江仙琚制药股份有限公司生产;盐酸利多卡因注射液(国药准字H37023473)为山东华鲁制药有限公司生产。②实验分组及给药:采用随机数字法将动物分为4组:生理盐水组8只,给予生理盐水2mL灌胃和局部注射生理盐水1mL/kg;环丙沙星组10只,给予盐酸环丙沙星800mg/kg;强的松龙组10只,给予1:2醋酸强的松龙注射液和盐酸利多卡因注射液,1mL/kg。联合作用组12只,给予盐酸环丙沙星800mg/kg和强的松龙1mL/kg。各组给药1次,d。⑨实验评估:分别于给药后2周、4周对实验动物进行体位、步态的行为学观察,局部组织病理学检查及流式细胞仪检测。实验过程对动物的处理符合动物伦理学标准。结果:40只大鼠全部进入结果分析。①行为学观察:用药后联合作用组大鼠跛行出现最早,环丙沙星组、强的松龙组次之,生理盐水组未出现。②大体标本观察:环丙沙星组、强的松龙组、联合作用组大鼠跟腱不同程度变为灰褐色,无定形外观肌组织。③光镜和电镜检测:与生理盐水组比较,其他3组大鼠跟腱损伤表现加重。④流式细胞仪检测:与生理盐水组比较,其他3组大鼠跟腱组织细胞凋亡率高(P〈0.01),并且联合组高于其他两组(P〈0.05),3组4周凋亡率高于2周(P〈0.01)。结论:氟喹诺酮类及糖皮质激素类药物应用后均对跟腱细胞有损伤�AIM: To investigate the influence of fluoroquinolone antibiotics and glucocorticosteroid on the Achilles tendon injury in rats, and determine if they have synergistic effect. METHODS: The experiment was carried out in the test animal center of Gansu College of Traditional Chinese Medicine from August to September in 2007. Forty SPF adult Wistar rats of either gender and weighing 200-250 g were used in this study. Ciprofloxacin (Cipro) tablets were produced by Harbin Pharmaceutical Group Co.,Ltd (Licence number: H.M.L.N. H23022275); prednisolone acetate (Pred) injection was produced by Zhejiang Xianju Pharmaceutical Co.,Ltd (Licence number: 1996-185301); lidocaine hydrochloride (LIDHC1) injection was produced by Shandong Hualu Pharmaceutical Co.,Ltd (Licence number: H.M.L.N. H37023473). Forty rats were divided randomly into four groups: Sodium chloride group (n=8): Intragastric administration of 2 mL and local injection of 1 mL/kg were given to every rat. Cipro group (n=10): Intragastric administration of 800 mg/kg Cipro was given. Pred group (n=10): Injection of Pred and LIDHC1 (1 mL/kg) according to the ratio of 1 : 2 was given. Joint action group (n=12): Cipro (800 mg/kg) associated with Pred (1 mL/kg) were given. All groups were treated one time every day. The observation of animal ethology such as posture and gait, histopathological examination of local tissues and flow cytometry detection were performed at 2 and 4 weeks after operation, respectively in the experimental animals. All the disposals were in accordance with the guideline of animal ethics. RESULTS: Forty rats were all involved in the result analysis. Animal ethology: Claudication occurred the earliest in the joint action group, then Cipro group and Pred group. No claudication was found in sodium chloride group.
