脾细胞和肝脏非实质细胞门静脉输注诱导免疫耐受的研究

摘  要:目的研究门静脉输注供体脾细胞和肝脏非实质细胞对大鼠皮肤移植的影响。方法建立大鼠皮肤移植模型,实验共分4组:对照组,实验1、2、3组。各实验组于皮肤移植前门静脉输注供体脾细胞、肝脏非实质细胞及两种细胞联合输注。观察移植皮片存活时间,测定迟发型超敏反应和体外抗体形成细胞水平,检测白细胞介素-2(IL-2)和细胞黏附因子-1(ICAM-1)。结果各实验组移植皮片存活时间延长,各检测项目指标较低,与对照组比较差异有显著性(P<0.05),以联合输注组为著。两种细胞单独输注组的存活时间差异无显著性(P>0.05)。结论皮肤移植前门静脉单独或联合输注两种细胞可延长移植物的存活时间,诱导免疫耐受,以联合输注组的作用最显著。其机制可能与降低细胞及体液移植免疫反应,IL-2、ICAM-1的表达有关。Objective To research the effects of donor spleen cells and liver nonparen chymal cells transplantation via portal vein on rat skin graft. Methods After the establishment of skin transplantation model in rats, the recipients were randomly calssified into 4 groups. The control group were given no treatment. Group Ⅰ and Group Ⅱ were given donor spleen cells and liver nonparenchymal ceils via portal vein before skin transplantation, respectively. Group Ⅲ were treated with both of donor spleen ceils and liver nonparenchymal cells. The grafts survival time was then monitored. The delayed hypersensitivity, exoteric antibody forming cell,interleukin-2 and the cellular adhesion molecule-1 were assessed after cells reansplantation. Results The graft survival time in the treated groups was significantly longer than the one in the control group( P 〈 0.05 ) ; all the testing items in the treated groups especially in Group Ⅲ were significantly (P 〈 0.05) decreased. However, there was no significant diffenernce between Group Ⅰ and Group Ⅱ in graft survival time. Conclusion Transplantation of one or two kinds of cells can prolong graft survival time and induce immunotolerance in rats skin-graft model. And the combined injection of cells can get the best effect. The mechanisms may be associated with the depressed cellular and humoral transplantation immunity and reduced expressions of interleukin-2 and cellular adhesion molecule-1.
