摘  要:目的:研究左旋四氢巴马丁(l-THP)对TNF-α诱导的人脐静脉内皮细胞(HUVECs)与单核细胞THP-1的黏附作用及对血管细胞黏附分子-1(VCAM-1)表达的影响,探讨l-THP在抗动脉粥样硬化方面的作用。方法:以TNF-α刺激体外培养的HU-VECs细胞,加入不同浓度的l-THP处理,测定HUVECs与单核细胞THP-1的黏附能力,并用酶联免疫吸附实验(ELISA)及西点法(western blotting)测定血管细胞黏附分子-1的表达。结果:l-THP(30μM)可以明显抑制TNF-α诱导的HUVECs与THP-1的细胞间黏附作用,其黏附率仅为TNF-α组的70.37±1.64%;ELISA实验中30μM浓度的抑制率为38.36±0.5%;western blotting实验同样显示l-THP(30μM)能显著降低VCAM-1的表达。结论:l-THP能够抑制内皮细胞与单核细胞的黏附及VCAM-1的表达,提示其在抗动脉粥样硬化作用中具有潜在的价值。Objective:To determine the effect of pharmacological doses of l-THP(30 μLM, 10μM and 3 μM) upon the adhesion of monocytic THP- 1 cells to human umbilical vein endothelial cells ( HUVECs ) , as well as the expression of vascular cell adhesion molecule ( VCAM-1 ). Methods: HUVECs were cultured in 96-well plate, pretreated with different concentrations of 1-THP, TNF-α was used to induce inflammatory responses in HUVECs. Then, HUVECs were co-incubated with THP-1 cells, the adhesion percentage is measured by multilable counter. The expression of vascular cell adhesion molecule ( VCAM-1 ) was evaluated by western blotting and ELISA. Results: 30 μM of l-THP-treatment inhibited the TNF-α induced expression VCAM-1 and THP-1 cells adhesion to HUVECs. Conclusion: Our results suggest that 1-THP inhibits the expression of molecules involved in the inflammatory process. Therefore, it could be used as an alternative in the treatment of chronic inflammatory diseases such as atheroselerasis.
