摘 要:目的:探讨运动环境对低浓度铅暴露小鼠记忆能力的影响和机制。方法:离乳健康ICR雄性小鼠随机分为实验组(铅暴露)和对照组,每组分别采用一般饲养(n=10)和一般饲养加运动(n=10),实验组采用铅浓度为0.2%饮用水进行低浓度铅暴露8周;利用原子吸收分光光度记配套石墨炉方法测定血铅和海马铅水平;Morris水迷宫实验法检测空间记忆能力;免疫组化法观察组织学改变。结果:各实验组血铅和海马铅浓度显著高于对照组(P<0.01);海马铅浓度实验组组间比较,一般饲育高于一般饲养加运动(P<0.05)。空间记忆实验中,达到避难平台的潜伏时间实验组长于对照组(P<0.05);实验组组间比较,一般饲养组长于一般饲养加运动(P<0.05);病理组织学结果发现,实验组一般饲养可观察到局灶性坏死,一般饲养加运动只表现为神经细胞局部分布不均和变性、其受损伤程度为轻度。结论:运动环境的刺激可能对空间学习能力的改善和脑海马组织的可塑性及代偿性的发挥起到一定的促进作用。Objective To explore the effects of motion environment on memory capacity in low-level lead exposure mice and mechanism. Methods Healthy weaned male ICR mice were randomly divided into control group and experimental group (lead exposure group) and then every group was divided into 2 groups: common fceding group and common fceding plus motion group (n= 10). The mice in experimental group were fed with 0.2% low-level lead drinking water for 8 weeks. Luminosity test (using an atom to absorb a beam split), Morris water maze test and immune test were applied respectively to detect the levels of blood lead and hippocampus lead and the impact o1 lead on mouse space memory and the histological changes. Results The lead levels in blood and hippocampus in experimental group were significantly higher than those in control group (P〈0. 01). The lead level in hippocampus in common fceding group of experimental group was higher than that in common fceding plus motion group (P〈0. 05). In space memory test, the latent time to reach refuge platform in common fceding group of experimental group was slower than that in control group (P〈0.05), and it was higher in common fceding group of experimental group than that in common fceding plus motion group (P〈0.05). The pathohistological results showed that focal necrosis was found in common fceding groups of experimental group, and focal uneven distribution, degeneration of never cells were found in common fceding plus motion group of experimental group. Oonclusion The stimulation of motion environment can improve the ability of space memory and promote the hippocampus plasticity and compensatory in low-level lead exposure mice.
