摘  要:为研究fps基因在烟草发状根中的表达对发状根生长及物质代谢的影响,对fps转基因发状根株系k-3离体培养条件下的生长特性及抗菌性进行了研究.在摇瓶悬浮培养过程中,发状根生长呈现"S"型曲线,最初生长缓慢,到第10天开始迅速生长,25 d进入生长平台期;利用气升式内环流生物器,在25℃通气量为2 L/min,接种量为1 g的条件下培养25 d,发状根鲜重达94.7 g/L;以赤星病菌(Alternaria alternata)粗毒素为诱导子对发状根诱导培养,转基因发状根浸提液在黑胫病根刺接种试验中,能够明显提高烟草幼苗对黑胫病的抗性.To study the effects of Famesyl Diphosphate Synthase gene (fps) expressing in tobacco hairy root on its growth and metabolites, transgenic hairy root strain k-3, expressing Mentha fps was cultured in vitro in two ways: suspension culture and bioreactor. It was found that the maximal production of hairy root as 94.7g/L was obtained in a internal loop air lift bioreactor when 1 g hairy roots cultured in 2 L liquid 1/2 MS medium for 25 d under 25 ℃ in dark condition. After induced by elicitor produced from Alternaria alternata, some fungi-resistance substance could accumulate in fps transgenic hairy root. The juice extracted from these hairy roots could prevent tobacco seedling from attacking by Phytophthoro parasitica. This result provides a bioreactor way to produce fungi inhibitor efficiently through transgenic hairy roots culture in vitro.
