调肝理脾方对酒精性大鼠肝纤维化的抑制作用及其方证探讨

摘　　要：目的:探讨调肝理脾方对酒精性大鼠肝纤维化的抑制作用。方法:Wistar雄性大鼠以"酒精-吡唑-玉米油"混合液灌胃16周,制备酒精性肝纤维化大鼠模型,将存活动物随机分成3组:模型对照组、调肝理脾方治疗组、西药对照(安珐特)治疗组,另设正常对照组。4周后检测大鼠血清中肝纤维化指标,观测肝组织病理学及肝组织中TGFβ-1蛋白表达变化。结果:16周末模型大鼠肝功能显著异常,肝纤维化明显;经4周治疗,与模型组比较,治疗组及对照组大鼠血清中HA含量均有显著下降趋势(P<0.01),且调肝理脾方能显著下调大鼠肝组织中TGFβ-1蛋白含量,其效果优于西药对照组。结论:调肝理脾方能有效阻止和逆转酒精性肝纤维化的进程,疗效优于西药对照组,方证相关有其病理学基础。Objective： To probe the inhibiting action of Tiaoganlipi prescription on alcohol hepatic fibrosis in rats. Methods： Wister male rats were administered by intragastrical peffusion by some mixture of ＂alcohol-Pyrazole-eoil oil＂ for sixteen weeks. And they were randomly assigned to three groups, a model control group, a Tiaoganlipi prescription group, a positive control group. After four weeks＇ intervention, To observation variety of hepatic fiborsis index, liver tissue pathology and expression of TGF-β1 in liver tissue. Results： After sixteen weeks, the model rats＇ hepatic functions were changed and abnormal, and fibrosis in hepatic histopathology. When giving intervention for four weeks, HA＇ s content had a downtrend （ P 〈 0. 01 ） in rats blood serum of the Tiaoganlipi prescription group, the positive control group compared with them of model group, and expression of TGF-β1 is depressed in Tiaoganlipi prescription group compared with positive control group. Conclusion： Tiaoganlipi prescription had an active effect on inhibiting of reversing of alcohol hepatic fibrosis in rats, and the conelativity of prescription with syndrome has pathological basis.
