摘 要:目的探讨用固相萃取-高效液相色谱(SPE-HPLC)法测定甘草中甘草苷和甘草酸含量。方法采用正交试验法考察真空度、上样量、洗脱剂用量和洗脱流速对固相萃取工艺的影响,用高效液相色谱梯度洗脱法测定各萃取液中甘草苷和甘草酸含量。结果甘草最佳固相萃取条件为真空度0.01MPa,上样量1.5mL,洗脱剂用量25mL,洗脱速率3mL/min;甘草苷和甘草酸的方法回收率分别为100.85%和99.67%。结论SPE-HPLC法操作简单,准确可靠,可用于甘草及含甘草的复方制剂中甘草苷和甘草酸含量测定。Objective To study the detection method for liquiritin and glyeyrrhizic acid in Radix Glycyrrhizae by solid phase extraction (SPE) and HPLC. Methods The orthogonal design was used to investigate the effect of four factors, including vacuum tightness, sample size, amount of eluent and eluting rate, on the solid phase extract technology. Liquiritin and the glycyrrhizie acid in various extract solutions were detected by gradient elution and HPLC. Results The optimized conditions of solid phase extract were as follows: the vacuum tightness was 0. 01 MPa, the sample size was 1.5 mL, the amount of eluent was 25 mL and eluting rate was 3.0 mL/min, The recovery rate of liquiritin and glycyrrhizie acid was 100.85% and 99.67% respectively. Conclusion The established method is simple, reliable and suitable for the determination of liquiritin and glyeyrrhizie acid in Radix Glyeyrrhizae and compound preparations containing glycyrrhiza.
