异丙酚靶控输注系统研究不同麻醉深度下兔脑c-fos mRNA水平的改变

摘  要:目的:研究异丙酚不同麻醉深度对兔脑c-fos mRNA水平的影响。方法:30只日本大耳兔,随机分为对照组、浅麻醉组、深麻醉组,每组各10只。应用异丙酚靶控输注系统,研究异丙酚不同麻醉深度下大脑额叶皮质,小脑皮质浦肯野细胞层、颗粒细胞层c-fos mRNA表达水平的改变。结果:与对照组比较,大脑额叶皮质,小脑皮质浦肯野细胞层c-fos mRNA表达水平随着麻醉深度的增加而显著升高(P<0.05);小脑皮质颗粒细胞层c-fos mRNA表达水平在麻醉状态下显著升高(P<0.05),但麻醉深度的增加并不能进一步降低其灰度值。结论:异丙酚通过增加c-fos mRNA的表达,影响随后反应基因的转录而发挥抑制中枢的作用。异丙酚抑制中枢的作用部位存在区域性差异。Objective:To research the alterations of c-fos mRNA in discrete brain regions of rabbit during propofol anesthesia. Methods:30 rabbits were randomly divided into three groups with ten each :controll group,light anesthetic group ,deep anesthetic group. By using target-controlled infusion of propofol,the levels of c-fos mRNA in frontal cortex (FC),cerebellar purkinje cells layer (PCL),cerebellar granule cortex layer (GCL) were observed under different anesthetic depths. Results :With the increase of anesthetic depth,the levels of c-fos mRNA in FC and PCL increased significantly compared with controll group(P〈0.05), The levels of c-fos mRNA in GCL increased significantly in both anesthetic groups(P^0.05) ,but the grayscale scores were not decreased further with the increase of anesthetic depth. Conclusions :Propofol exerts its anesthetic action through increasing the expression of c-fos mRNA,subsequently working on fos-related gene transcription. This kind of influence differes in different brain regions.
