摘 要:目的揭示川乌煎煮时间、给药剂量与毒性的相关性。方法采用均匀设计的方法,将川乌分别煎煮15 m in,30m in,1,2,3,4,6 h,生药含量依次为临床用量的6,12,24,48,72,96,120倍。模型对照组和各给药组于复制痹证动物模型后,按组给药30 d。观测动物生长发育、血液学、血液生化学、脏器系数、病理组织学变化及停药15 d后上述指标的变化。将结果进行多元回归分析,得出川乌煎煮时间、剂量与毒性的相关性。结果川乌水煎液能抑制痹证动物的足跖肿胀;对模型动物的生长发育无明显影响;对其造血系统无明显毒性;对其肝脏、肾脏、心脏有一定毒性。川乌水煎液毒性最大的煎煮时间和给药剂量是30 m in,120倍。结论煎煮30 m in,给药剂量为临床用量120倍时,川乌水煎液对痹证动物产生的毒性最大,但药效最佳。Objective To study the correlation between decocting time (DT), administration dosage(AD) and the toxicity of Radix Aconiti(RA). Methods According to uniform designed technology, RA was decocted with different duration of 15 minutes, 30 minutes, 1 hour, 2 hours, 3 hours, 4 hours and 6 hours,and the content of RA was 6 times, 12 times, 24 times, 48 times, 72 times, 96 times, 120 times of clinic AD respectively. All rats except normal control group were made up BiZheng animal model, and administered RA decocted liquids for 30 days. The changes of hematology, blood biochemistry, organ coefficient, histopathology test index were detected before and after 15 days. The regression analysis was adopted to study the relativity between DT, AD and the toxicity of RA. Results RA decocted liquids could decrease voix pedis tumefaction of BiZheng model animals, but had no effect on the growth and development and hematopoietic system of model animals. They had certain toxicity to livers, kidneys, and hearts of the model animal. The DT and AD of maximal toxicity of RA decocted liquids were 30 minutes and 120 times. Conclusion RA decocted liquids with DT 30 min and AD 120 times have the maximal toxicity and the best medicinal effect.
