摘 要:目的初步筛选补骨脂平喘有效部位。方法建立大鼠哮喘模型,采用ELISA法检测补骨脂不同溶剂提取物对血清IFN-γ/IL-4/IL-25含量的影响,筛选补骨脂平喘有效部位。结果①对IL-5含量的影响:醇提浸膏、醇提水萃取物、水提取物和醇提甲醇洗脱物对血清IL-5含量抑制明显,而醇提石油醚萃取物、醇提醋酸乙酯萃取物抑制作用不明显。②对IL-4含量的影响:醇提浸膏、醇提石油醚萃取物和水提物对血清IL-4含量有较强的升高作用,而醇提醋酸乙酯萃取物、醇提水萃取物和醇提甲醇洗脱物作用不明显。③对IIFN-γ含量的影响:醇提浸膏和醇提石油醚萃取物明显提高IFN-γ含量,而醇提醋酸乙酯萃取物、醇提水萃取物和醇提甲醇洗脱物却降低大鼠体内血清IIFN-γ含量,水提取物对IIFN-γ含量影响不明显。结论补骨脂乙醇提取物浸膏、石油醚萃取物和甲醇洗脱物是补骨脂平喘主要有效部位。Objective To screen the antiasthmatic active constituents of Psoralea Fruits. Methods After animal model in rats was established, the concentration of IFN-γ, IL-d, IL-5 in serum were detected by ELISA after each solvent extraction being administered to rats. Then the antiasthmatic active constituents of psoralea fruits was screened according to the effect. Results (1) About IL - 5 : compared with the normal group, the contents of IL - 5 in serum were significantly restrained in the following groups including the ethanol - extracted extractum group, ethanol - extracted H2O-abstraction group, H2O-extracted extractum group, and ethanol- extracted methanol-eluate group, respectively; while not significantly in ethanol -extracted iigarine- abstraction group and ethanol- extracted acetoacetate- abstraction group, respectively. (2)About IL-4: compared with the normal group,the contents of IL-4 in serum were significantly increased in ethanol - extracted extractum group, ethanol - extracted iigarine - abstraction group and H2O-extracted extractum group, respectively; while not significantly in ethanol-extracted acetoacetate- abstraction group, ethanol- extracted H2O-abstraction group and ethanol -extracted methanol - eluate group, respectively. (3)A- bout IFN - γ, compared with the normal group, the contents of IFN - γ in serum were significantly increased in following groups including ethanol - extracted extractum group, ethanol - extracted iigarine - abstraction group, respectively; while were decreased in ethanol -extracted acetoacetate - abstraction group, ethanol -extracted H2O -abstraction group, ethanol -extracted methanol - eiuate group, and no significant change in H2O -extracted extractum group. Conclusion Ethanol -extractum, iigarine- abstraction and methanol - eluate are mainly antiasthmatic active constituents of psoralea fruits.
