摘 要:目的建立黄金咽喉片中柠檬酸的薄层鉴别及含量测定方法。方法采用薄层色谱法对柠檬酸进行定性鉴别,并采用高效液相色谱(HPLC)法测定柠檬酸的含量,色谱柱为:Dikma Kromasil C18柱(250mm×4.6mm,5μm);流动相为,乙腈-0.1%磷酸0.1%三乙胺溶液(1:99);检测波长:210nm;柱温:25℃;流速:0.6ml·min^-1;进样量:10μl。结果在薄层层析色谱中能检出柠檬酸,柠檬酸在1.04~10.4μg范围内具有良好的线性关系(r=0.9999),平均回收率为98.85%,RSD为0.37%。结论该方法简便、快速、有效、灵敏、准确,具有良好的重复性和回收率,可作为该制剂中柠檬酸的定性与定量分析方法。Objective To establish the method for the TLC identification and content determination of citric acid in Huangjin Yanhou Tablet. Methods Citric acid in Huangjin Yanhou Tablet was identified by TLC, and the content of citric acid was detected by HPLC. The separation was carried out on a Dikma Kromasil C18 column(250 mm ×4.6mm,5 μm). The mobile phase was acetonitrile -0.1% phosphoric acid 0.1% triethylamine solution( 1: 99). The detection wavelength was set at 210 nm,the column temperature was 25℃ ,the flow rate was 0.6 ml · min^-1 and the sample size was 10 μl. Results Citric acid could be detected by TLC, the calibration curve showed good linearity over the range of 1.04 - 10.4 μg( r = 0. 999 9 ). The average recovery was 98. 85% with RSD 0. 37% . Conclusion This method is simple, rapid, efficient, sensitive, accurate and with good repeatability and recovery,it can be used as a quantitative analysis method for this preparation.
